| BackgroundThe most of clinical observations suggest that in patients with cirrhosis associated with different levels of intestinal endotoxemia. In liver damage, the injury of intestirral mucosal barrier can not effectively remove endotoxin, and the formation of intestinal endotoxemia. It is rarely repoted that acute liver injury whether associated with intestinal endotoxemia. A large number of studies have confirmed that Bifico can improve chronic liver damage imbalance of intestinal flora, to reduce intestinal endotoxemia, It is unclear that bifico whether have a protective effect to acute liver injury intestinal endotoxemia. In order to clarify this issue, the design of this topic.ObjectiveTo identify the protective effects of Probiotics on intestinal endotoxemia in rats with Acute liver injury.MethodsThe model of acute liver injury was induced by subcutaneous injections of thioacetamide (TAA).40SD rats were randomly divided into five groups:normal control group (N), model group (T), zheng chang sheng group(Lã€H) and bifid triple viable group (P). The levels of endotoxinã€ALT(alanine aminotransferase)ã€AST(aspartate aminotransferase)ã€LDH(lactate dehydrogenase)ã€MDA(malondialdeyde) were measured using biochemical method, histopathology of liver(HE staining) were observed, and hepatocyte apoptosis was detected by DUTP nick en labeling (TUNEL).Results(1) In comparison with those in model group, focal necrosisi and infiltration of inflammatory cell were appeared and the hepatic apoptosis index were improved significantly. The liver damage were lesser in group Lã€Hã€P than in model group.(2) The serum levels of endotoxin were significantly increased in model group as compared with those in normal control group (endotoxin:0.086±0.02vs0.34±0.07,P<0.01). In comparison with those in model group, the tissue expression of endotoxin were significant decrease in group Lã€Hã€P (endotoxin:0.16±0.05vs0.34±0.07,0.09±0.05vs0.34±0.07,0.11±0.04vs0.34±0.07,P<0.01)(3) The serum levels of ALTã€ASTã€LDHã€MDA were significantly increased in model group as compared with those in normal control group (ALT:5.99±2.21vs47.05±13.06, P<0.01; AST:18.42±5.28vs46.93±42.93, P<0.01; LDH:2555.69±622.19vs 4443.07±605.68,P<0.01:MDA:3.48±0.63vs5.40±1.26,P<0.01).In comparison with those in model group, the tissue expression of ALTã€AST were decrease in group L.H. P (ALT:32.45±17.4vs47.05±13.06,29.35±12.31vs47.05±13.06,30.08±17.84vs47.05±13.06,P>0.05;AST:34.73±6.02vs46.93±42.93,35.83±7.54vs46.93±42.93,31.63±12.46vs46.93±42.93,P>0.05;LDH:4295.79±780.9vs4443.07±605.68,3804.46±674.65vs4443.07±605.68,4142.33±753.2vs4443.07±605.68,P>0.05;MDA:5.08±1.15vs5.40±1.26,5.12±0.91vs5.40±1.26,4.82±0.73vs5.40±1.26,P>0.05)(4) The tunel staining analysi s:More TUNEL positive cell s were found in model group than normal group and group L.H.P(34.13%±12.59ï¼…vs3.31%±2.23,35.71%±12.87ï¼…vs3.10%±2.03ï¼…,P<0.01;11.14%±6.04ï¼…vs34.13%±12.59ï¼…,8.35%±5.58ï¼…vs34.13%±12.59ï¼…,9.89%±5.12ï¼…vs34.13%±12.59ï¼…,P<0.01:10.23%±6.60ï¼…vs35.71%±12.87ï¼…,8.84%±5.09ï¼…vs35.71%±12.87ï¼…,9.93%±5.79ï¼…vs35.71%±12.87ï¼…, P<0.01)Conclusion(1) The rats with acute liver injury caused by TAA have intestinal endotoxemia.(2) Probiotics protects rats f.rom acute liver injury by improving intestinal dysbacteriosis and reducing the level of plasma endotoxin. |
