Objective: To compare the serum Klotho protein,NO and ET-1levels inpatients with varying degree of hypertension and to explore the role andrelativity of Klotho protein,NO, ET-1in the development of hypertension.Methods:126persons that chosen from patients hospitalized inBethune’s The First Hospital of Jilin University Cardiovascular center andphysical examination center from2011-8to2011-11, aged from18to75,excluding those with acute and chronic liver and kidney disease, cancer,coronary atherosclerotic heart disease, cerebrovascular disease, peripheralvascular disease, autoimmune disease, infectious disases, and secondaryhypertension,were divided into4groups by blood pressure levels: controlgroup (n=30), hypertension grade1group (31cases), hypertension grade2group (33cases), hypertension grade3group (32cases). Collect3to5mlblood from all patients early in the morning before breakfast then centrifugatedin speed of3000rev/min for10-15min, and the supernatant were saved in-80°C refrigerator for test. Serum Klotho protein, ET-1levels were detected byELISA. NO levels were detected by Nitale redurtase. Glucose, cholesterol,triglycerides, low-density lipoprotein cholesterol, High density lipoproteincholesterol levels were detected by applying automatic biochemistry analyzer.Results:1. With increased blood pressure, serum Klotho protein levelgradually decrease, which were negatively correlated with blood pressure levels.Serum Klotho protein is no significant difference between hypertension grade1group and control group. There is significant difference between any of othergroups (P <0.05).2. With increased blood pressure, serum NO level graduallydecrease, which were negatively correlated with blood pressure levels. There is significant differences between any of two groups (P <0.05).3. With increasedblood pressure, the expression of ET-1was gradually increased, which waspositively correlated with blood pressure levels.There is no significantdifference between hypertension grade1group and control group, betweenhypertension grade2group and hypertension grade1group, betweenhypertension grade3group and hypertension grade2group. There issignificant difference between any of other groups (P <0.05).4. The serumlevels of Klotho protein was positively correlated with NO, no correlationwith ET-1. The level of NO was negatively correlated with ET-1.5. SerumKlotho protein were negatively correlated with cholesterol, Low-densitylipoprotein cholesterol (LDLC), has no correlation between glucose,triglyceride, High density lipoprotein cholesterol (HDLC).Conclusion:1. With increased blood pressure, serum Klotho protein andNO level gradually decrease, ET-1content gradually increased.2. Klothoprotein is decrease compired with the control group in the patients sufferedfrom hypertension,and in the negative related with hypertension grades,so weconsider that one of the causes of hypertension may be lack of endogenousprotein Klotho.3. There is a positive correlation between Klotho protein andNO,and thay have a negative related with hypertension grades,so it give usguiding significance in the future with prevention and treatment of high bloodpressure and evaluating the prognosis.4. Serum Klotho protein may have nocorrelation with ET-1.
