| Background Benign prostatic hyperplasia and prostatitis is the most common benign disease in the two male genitourinary tract. Many studies have found that often exist in a middle-aged men, but the association between domestic and foreign scholars present there is no unified understanding.To detect the expression of IL-8FGF-2and MMP-2in benign prostatic hyperplasia(BPH)and BPH combined with prostatitis, and to evaluate the effect of inflammation on the development and progression of BPH.Objective In many reports, IL-8, FGF-2and MMP-2participated the process of prostatitis and benign prostatic hyperplasia, but they were less report in the role of prostatitis participated benign prostatic hyperplasia. To detect the expression of IL-8〠FGF-2and MMP-2in benign prostatic hyperplasia(BPH)and BPH combined with prostatitis, and to evaluate the effect of inflammation on the development and progression of BPH.Methods63specimens were obtained from patients undergoing transurethral prostatic resection(TURP). The paraffin section of the specimens were stained with hematoxylin and eosin,and observed to examine the inflammation pathological changes. Depending on whether the benign prostatic hyperplnsia combined with prostatitis, those specimens were divided into group of benign prostatic hyperplnsia(A group)and group of benign prostatic hyperplnsia combined with prostatitis(B group), using immunohistochemistry and Real—time fluorescence quantitative PCR(RT—PCR)tO dealt the expressions. The relevant data were processed with statistics method.Rssults1.HE dyeing results In63cases of prostate hyperplasia with prostatitis person for41cases(65.8%) and with22cases for benign prostatic hyperplasia(35.2%).In41cases combined with prostatitis,there were different degree of inflammatory cells embellish.2. Immunohistochemistry results IL-8, FGF-2and MMP-2were all expressed in group A and group B. The optical density of IL-8in group A wasχ±s=0.2167±0.0391,while the optical density of IL-8in group B was χ±s=0.2398±0.0401.The group B of tissues were significantly higher than group A of tissues,and the difference were significant (P=0.038<0.05).The optical density of FGF-2in group A was χ±s=0.2917±0.0283,,while the optical density of FGF-2in group B was χ±s=0.3108±0.0357.The group B of tissues were significantly higher than group A of tissues,and the difference were significant (P=0.026<0.05). The optical density of MMP-2in group A was χ±s=0.3221±0.0283,,while the optical density of MMP-2in group B was χ±s=0.4018±0.0371.The group B of tissues were significantly higher than group A of tissues,and the difference were significant (P=0.014<0.05).3. RT—CR results IL-8, FGF-2and MMP-2mRNA were all expressed in group A and group B. The relative ratio of IL-8, FGF-2and MMP-2mRNA in group A is0.056±0.015ã€.041±0.012ã€0.107±0.023. The relative ratio of IL-8, FGF-2and MMP-2mRNA in group B is0.181±0.03ã€0.075±0.011ã€0.231±0.062.The group B of tissues were significantly higher than group A of tissues,and the difference were significant (P<0.05).Conclusion IL-8〠FGF-2and MMP-2in the tissues of benign prostatic hyperplnsia combined with prostatitis were significantly increased.
