| Objective:To observe the expression of nuclear factor kappa B (NF-kB) and macrophage-specific chemoattractant protein-1(MCP-1) in5/6Nephrectmized Rats. And to investigate part mechanism of action for Liuweidihuang Decoction on Renal tubulointerstitial fibrosis.Methods:The sixty male Sprapue-Dewley rats models were randomly divided into5groups(n=12):normal groupã€sham-operated groupã€model group〠enalapril group and Liuweidihuang Decoction group. The model group, enalapril group and Liuweidihuang Decoction group adopted5/6nephrectomized induced into renal failure model.The sham-operated group, the same time the two operations, each time only to open the abdominal cavity, the abdomen was closed after the exposure to kidney.7days after the second surgery, the rats were treated by gavage. Rats were sacrificed eight weeks’ later. To study the efects of Liuwei dihuang Decoction on the functions and pathologic changes by HE staining and Masson staining of remnant kidney.Immunohistochemistry was used to measure the expressions of NF-κB and MCP-1in nephridial tissue.Results:Firstly, Duplicating processes of5/6Nephrectmized model was successful by oberserving tubulointerstitial histomorphology with HEã€Masson staining.The degree of renal interstitial fibrosis in the Liuweidihuang Decoction group and the enalapril group was much slighter than that in the model group; Secondly, The expression of NF-κB and MCP-1in the Liuweidihuang Decoctior group was lower than the model group. Thirdly, the Liuweidihuang Decoctior group can significantly decrease the serum levels of BUNã€Scr and urinary proteir leakage。Conclusion:Liuweidihuang Decoction may inhibit the abnormal activation of NF-κB, reduce the expression of MCP-1and other inflammatory factors and decrease the formation of Collagen â…¢ and other extracellular matrix, so as tc reduce the inflammation and fibrosis and delay the progress of renal interstitial fibrosis。... |
