The Experimental Study On Effects Of Liuweidihuang Decoction On VEGF And TNF-α Expressions With5/6Nephrectomized Rats

Objective:Observing Liuweidihuang decoction’s influences on ex-pressions of Vascular Endothelial Growth Factor(VEGF) and Tumor Necrosis Factor-a(TNF-a) in5/6nephrectomized rats remnant kidney, to discuss the mechanisms which Liuweidihuang decoction may use to protect renal function and delay kidney damage.Methods:72SD rats were randomly divided into6groups:normal group, sham operation group, and4operation groups,12rats in each group. The operation groups were performed a surgery of5/6nephrectomy induced chronic renal failure rat model, and meanwhile sham operation group undergoing sham operations:only open abdomen and exposed kidneys, ending with abdominal closure.3days after second operation, The43survivals in surgical group were, according to random number table, divided into3groups:Liuweidihuang decoction group, enalapril group and model group. All rats were treated by gavage and put to death after eight weeks, then observe morphological changes of remnant kidney and the expression of VEGF and TNF-a assayed by immunohistochemistry staining.Results:1.Observing remnant kidney’s morphology changes of5/6nephrectomy rats by HE staining and Masson staining, it turns out that CRF rat models induced by5/6nephrectomy were successfully reproduced, and that rats renal fibrosis degree can be reduced by Liuweidihuang decoction and enalapril;2.Semi-quantitative immuno-histochemical analysis of remnant kidneys belong to Liuweidihuang decoction group rats revealed that expressions of VEGF and TNF-awere less than remnant kidneys of model group;3. Liuweidihuang decoction can lower content of BUN and Scr, as well as24h urine protein quantity etc.Conclusion:Reducing the expressions of VEGF and TNF-α, by which Liuweidihuang decoction may relieve the inflammatory response and fibrosis, so as to protect the renal function.
