| The Hairy Anemone (Anemone tomentosa (Maxim.) Pei) is the buttercup family(Ranunculaceae) of silver lotus (Anemone) perennial herbs. The root of Hairy Anemonefor medicinal is used as clearing heat and removing toxicity, row pus, swelling bornmuscle scattered blood stasis, and the food products, cut anopheles, pesticides, such aseffectiveness. It wildly distributed in the southwest, northwest, northern and central ofChina. In order to improve the exploiting and using of Hairy Anemone resources andmedicinal value, the investigations on the chemical composition and bioactivities ofhave been carried out in this paper.The chemical composition of Hairy Anemone have been tested systematically bymeans of pre-test. It found that it contains flavonoids (2hydrogen flavonoids),coumarone, lactone, strong heart glucoside, anthraquinone and glucoside kind ofcomposition, saponins (saponins), sugar and glucoside, organic acid and phenoliccompounds component. There is no alkaloids, volatile components, amino acids,peptides, protein compounds.And then the experimental study of the biological activities for three kinds ofchemical composition has been taken:(1) Their antimicrobial activities in vitro wereevaluated by filter paper method. The results showed that the total flavonoids, saponinsand polysaccharide had no inhibitory effects to the two bacteria (Staphylococcus aureus,Eschercihia Coli).(2) MTT colorimetric assay was adopted to test their anti-tumoractivities to human hepatoma cell lines Hep-G2. The results showed that In addition tothe flavonoids, the total saponin and polysaccharides of Hairy Anemone both exihibitedno significant inhibitory effects on tumor cells.(3) Compared with Vc, hydroxylradicals and superoxide radical scavenging effect in the study, the results show that:Compared with Vc, the flavonoids, total saponins and total polysaccharides has strongantioxidant ability, has all concentration dependence relationship. Among them, theflavonoids of hydroxyl radical scavenging effect is stronger than Vc, superoxide anionto remove effects is weaker than Vc. And compared with the flavonoids, total saponinsand Vc, total polysaccharides antioxidant activity is the weakest.In view of flavonoids, total saponins and total polysaccharides had significantantioxidant and antitumor activity, according to the flavonoids, total saponins and totalpolysaccharides extracting rates, the extracting techniques were optimized by singlefactors and orthogonal design. The results indicated that the best extracting parameterswere as follows: the flavonoids extraction concentration of alcohol was60, the ultrasonic assisting time was80min, the ratio of solid to liquid was1︰50, theextractive times were3, the ultrasonic power was90W; the total saponins extractionconcentration of alcohol was60, the extracting temperature was75℃, the extractingtime was210min, the ratio of solid to liquid was1︰20; the total polysaccharidesextracting time was150min, the ratio of solid to liquid was1︰30, the extractive timeswere3, the extracting temperature was90℃.The growth inhibition was analysed by MTT the tumor of hepatocellular carcinomaHep-G2, SMMC-7721, human breast cancer cells MCF-7and people cervical cancercells Hela.The flavonoids were found to be active in effect on antitumours in vitro. Theconcentration was lmg/mL, the ratio of inhibition of tumor growth of hepatocellularcarcinoma Hep-G2, SMMC-7721, human breast cancer cells MCF-7and peoplecervical cancer cells Hela were above50%, and the IC50were0.5298mg/mLã€0.2047mg/mLã€0.2877mg/m1ã€0.2738mg/m1. The ratio of inhibition of cells has remarkabledose-dependence. Its anti-tumor activity in vivo was evaluated by H22cells-bearingmice models. The High(1.5g/kg)ã€middle(0.75g/kg) and low(0.375g/kg) dose flavonoidsof also significantly suppressed the tumor growth with tumor inhibition rate of38.57%ã€19.54%and13.92%respectively, also has remarkable dose-dependence. By comparingthe thymus and spleen index, found that flavonoids of Hairy Anemone can also reducethe spleen immune ability.This article lay the foundation research fire through to the chemical compositionsand biological activities for using of Hairy Anemone, for its antitumor drugdevelopment to provide the scientific data support, finding of activity screening with thefire in antitumor activity, but its antitumor activity at the beginning of the study and thefire grass at the beginning of the antitumor drugs research and development, and havegood development prospect. |
