Purpose: The constituents of Alpinia Roxb include the volatile oil,flavonoids,terpenoids,diphenylheptane,phenylpropanoids.Pharmacological studies show its activities ofanalgesic,sedative,anti-tumor,anti-oxidation,anti-bacterial,anti-thrombotic,anti-inflammatory,reducing blood press.Alpinia Nigra belong to Alpinia Roxb,which already has some reports about its medicinal value.However the systematical study on the chemical constituents and bioactivities is still scarce.In this paper,the dried branches and leaves of Alpinia Nigra are selected as the research object in order to consummate the medicinal plant parts of Alpinia Nigra and consummate the medicinal plant resources of Alpinia Roxb.Futhermore the chemical constituents and bioactivities are detremined so as to obtain potential lead compounds.Methods: The branches and leaves are extracted with 95% ethanol and recover ethanol to obtain the total extract.The appropriate proportion of distilled water is added into the total extract and then petroleum ether,ethyl acetate and n-butanol are used successively to obtain three parts with different polarity.MCI column chromatography,Silica gel column chromatography,Gel column chromatography and High performance liquid chromatography(HPLC)is used to separate and purify the chemical components,according to the physical and chemical properties and spectral data,the structures of the chemical components are analyzed and identified.Results: Fourteen chemical constituents are isolated from Alpinia nigra.They are identified as phytenal(1),?-tocopherol-quinone(2),galanal B(3),galanal A(4),coronarin B(5),pacovatinin C(6),p-hydroxycinnamic methyl ester(7),(-)-(7R,8S)-erythro-guaiacylglycerol(8),(-)-(7R,8R)-erythro-guaiacylglycerol(9),coniferaldehyde(10),vanillin(11),quercetin(12),quercitrin(13),isoquercitrin(14),Which are all isolated from the qplant for the first time.Conclusion: In this paper,the chemical components of the branches and leaves from Alpinia nigra are studied.Including the flavonoids,diterpenoids phenylpropanoids and other chemical components.The chemical composition of the leaves and branches of Alpinia nigra is preliminarily understood.At the same time,this study also tested the important biological activities of the isolated monomer compounds,such as anti-tumor,anti-bacterial,ion channel inhibition,anti-hepatitis b virus.When studying the cytotoxic activity of the monomer compounds on A549 cells(human non-small cell lung cancer cells)and MDA-MB-231 cells(human breast cancer cells),it is found that the inhibition rate of compound 4 with 10 ?M to A549 cells is 89.1%,while the inhibition rate of MDA-MB-231 cells is 54.8%.Compound 4 showes strong cytotoxic activity against A549 cells,which provides a preliminary clue for understanding the medicinal value of the leaves and branches of Alpinia nigra.
