Objective:In this study, rapid resolution liquid chromatography quadrupole time-of-flight massspectrometry (RRLC/Q-TOF-MS) was used to profile the metabolites of urine samples fromcancer patients and healthy controls. Multivariate statistical analysis method, that is, clusteranalysis, principle component analysis and partial least squares-discriminant analysis wereused to discriminate between the urine samples and find the biomarkers.Methods:1. Sample preparation: The samples were centrifuged at12000rpm for5min, the upperphase was filtered by0.45μm filter for LC-MS analysis2. LC-MS analysis of urine samples: The urine samples were analyzed byRRLC/Q-TOF-MS. The urine metabolic profile was obtained. Then find the differencesbetween them.3. Data analysis: Multivariate statistical analysis was used to analysis the data of urineand find biomarkers. And then judge the efficacy of the biomarkers to divide the urinesamples. Cluster analysis, principal component analysis (PCA) and partial leastsquares-discriminant analysis (PLS-DA) were performed for the multivariate statisticalanalysis.Results:1. In the research of colorectal cancer, the clear separation of two groups was observed inthe PCA scores plot where each point represents a sample. From the PCA loading plot,19compounds were found as biomarkers in both positive and negative ion mode.2. In the research of gastric cancer, the acquired metabonomic data was analyzed bymultivariate analysis. In the PCA scores plot, each group showed its own individual in-groupsimilarity, two groups were clearly distinguished from each other. We also found24 biomarkers from the PCA loading plot and the model based on these biomarkers showed thehigh showed high sensitivity and specificity.Conclusion:In this study, the LC-MS method was used for the metabonomic analysis of urinesamples. The LC-MS method provide comprehensive quantitative and qualitative information,in order to determine the differences between the groups based on the metabolite profile,multivariate statistical analysis was used in this study. The results of this study demonstratedthat the metabonomics is a powerful and useful tool for the analysis of the cancer. |
