Alfalfa is a legume (Leguminosae) perennial herb, alfalfa (Medicago.sativca Linn.) entire grass, for medicinal and edible plant, alfalfa contains a variety of natural effective components, has good medicinal value, taking the alfalfa as raw material, the active component of total flavonoids extraction, enrichment, purification of further on its pharmacological activity research, for the development of medicine for treatment of piles foundation.This paper firstly by using spectrophotometric method establishing alfalfa flavonoids content determination method of alfalfa, further optimization of the extraction technology of total flavonoids by macroporous adsorption resin, and method for optimizing the enrichment technology.Optimization of extraction technology of total flavonoids of alfalfa, method is by using the uniform design method to effects of flavonoids extracted factors optimization.The results show that, the optimal extraction process for solid/liquid ratio1:19, extracting for twice, extraction time128min, ethanol concentration of71%. This paper firstly by using spectrophotometric method establishing alfalfa flavonoids content determination method of alfalfa, further optimization of the extraction technology of total flavonoids by macroporous adsorption resin, and method for optimizing the enrichment technology.Total flavone of Alfalfa on the enrichment technology, used by macroporous resin adsorption were investigated, resin type, sample volume, elution volume, elution solvent and eluting velocity, the results show that, D101resin adsorption and desorption rate is higher, is ideal for alfalfa flavonoids purification resin types, and enrichment process optimal conditions for the sample volume,1:20, use the3Bv dosage of70%ethanol as eluting agent elution, eluting rate was2ml/0.79cm/.min.This article on alfalfa flavonoids were pharmacodynamic studies, in order to evaluate the alfalfa flavonoids in treatment of hemorrhoids, anti-inflammatory, analgesic, anti ulcer from several aspects, in vitro pharmacodynamic studies, first study showed that alfalfa flavonoids mice ear swelling caused by dimethylbenzene and egg white induced ear edema of mice and agar granuloma the effect of inflammation effect is obvious; the second study confirmed the drug analgesia in Physics (hot plate method), chemical analgesia (acetic acid writhing) have a significant role, finally by drilling method, tube two fold dilution methods were studied in vitro bacteriostasis, results showed that alfalfa flavonoids in the test dose of, E. coli, Staphylococcus aureus, Pseudomonas aeruginosa, Staphylococcus aureus has different degrees of inhibition, bacteriostasis effect is obvious; resistance to bleeding and blood coagulation research proof alfalfa flavonoids in the mice on bleeding time and blood coagulation time was significantly shorter, prove its to promote hemostasis and coagulation function, at the same time can be caused by acetic acid mouse anal exudates decreased, ulcer area narrowing, ulcer healing.Alfalfa extract flavones with cheap, easy to use, the advantages of fewer side effects, so it is an ideal drug in treating hemorrhoids, have better application prospect.
