| Traditional Chinese medicine Schisandra chinensis and Schisandra sphenanthera fruit are the dry fruit from Schisandraceae plant of Schisandra chinensis (Schisandra chinensis (Turcz.) Baill.) and Schisandra sphenanthera(Schisandra sphenanthera Rehd.et Wils) respectively.They have been widely used in clinical application with the functions of astringing, replenishing qi and producing body fluids, tonifying kidney and canning the mind. However, schisandra fruit from different origins have the remarkable difference in reducing enzyme activity of lignans, type and content of aspect, further more, the Schisandra quality is very unstable and they may made a direct impact on the clinical efficacy. Besides that, medicinal quality standard are lack of specificity. In this study, schisandra fruit from different origins have been discussed by the method of using HPLC and UPLC/Q-TOF-MS technology and have been made comparative study to distinguish the similarities and differences ignans chemical composition which included in Schisandra chinensis and Schisandra sphenanthera. Also, this study has set the control method of qualitative and quantitative, provide scientific basis for medicinal identification in order to ensure clinical use of drugs safety and effective. The main research results are as follows:First, through using the way of central composite design-response surface method to roptimizing the extraction technology of Schisandra fruit, taking the lignan concentration as indexes, receive the optimum extraction condition conditions as20times the volume of85%ethanol3times,170min. per time finally.Second, Schisandra chinensis and Schisandra sphenanthera from different areas have been used as the research object. In this easy, the simultaneous determination of Schisandra chinensis and Schisandra sphenanthera fingerprint have been set and the comprehensive analysis method have been used in the six main lignans concentration of the two schisandra, too. Besides that, the method have been used in medicinal material identification and source tracing of schisandra. The results showed that fingerprint’s similarity is greater than0.9, between Schisandra chinensis and Schisandra sphenanthera from different areas. But the Schisandra chinensis and Schisandra sphenanthera fingerprint similarity is less than0.35, it means that the fingerprint’s characteristic are strong and specificity. Some data shows that the quality volume fraction of Schisantherin A, Deoxyschisandrin in Schisandra sphenanthera are greater than Schisandra chinensis, but the quality volume fraction of Schisandrin, Schisandrol B, Schisandrin B from Schisandra chinensis are greater than Schisandra sphenanthera Also, lignans gross Schisandra chinensis is nearly two times than the Schisandra sphenanthera.Third, analysis the lignans from extract of schisandra chinensis by the method of UPLC/Q-TOF-MS. In positive ion mode, using the similarities and differences, which from mass spectrometric behaviour in order to carry on the comparative analysis on lignan components of the two.The result shows that the two have certain same chemical composition include that Schisandrin, Schisandrol B, Schisantherin A, Schisantherin B, Schisantherin C, Schisantherin D, Deoxyschisandrin. But they are different in concentration. The concentration of Schisandrin, Schisandrol B, Schisantherin D in Schisandra chinensis is high while the concentration of Schisantherin A, Schisantherin B, Schisantherin C Deoxyschisandrin in Schisandra sphenanthera is high. In addition, the unique elements of Schisandra chinensis are Gomisin H, y-Schisandrin, Schisandrin B, Schisandrin C, while the unique elements of Schisandra sphenanthera is M Kadsulignan M.
