Objective:Our aim is to find differentially expressed genes in hippocampus between the depressive disorder model rat with Gan-qi depression, normal control and rat treated by Shuyu capsule and Saikosaponin extract, which gives a preliminary discussion on the pathogenesis of depression with Gan-qi depression, as well as studies the effects of Shuyu capsule and Saikosaponin extract.Methods:After being chosen, depressive disorder model rat with Gan-qi depression were prepared by using chronicity mild stimulus (CMS) method. And then the rats were evaluated by sucrose preference test and open field test. Differentially expressed genes of amino acid neurotransmitter receptors in hippocampus between the depression model rat with Gan-qi depression, normal control and rat treated by Shuyu capsule and Saikosaponin extract were selected by the rat expression microarray analysis. Part of the amino acid neurotransmitter receptor gene expression were detected by Real-time PCR to test the accuracy of results.Results:Compared with the normal group, the total score of open field test of rats in depressive disorder with Gan-qi depression model group decreased obviously(P<0.01) and the sucrose intake of preference test decreased obviously(P<0.01). On the other hand, compared with the depressive disorder with Gan-qi depression model group, the open field scores of rats in Shuyu capsule, fluoxetine and Saikosaponin extract treatment groups increased remarkably (P<0.01, P<0.001, P<0.05). The sucrose intake of preference test of rats in Shuyu capsuleand fluoxetine treatment groups increased obviously(P <0.05, P<0.01), and there was no statistical differences in the two treatment groups(P>0.05). Though the sucrose intake of preference test of rats in saikosaponin extract treatment groups was no significant difference compared with the model rats, there is a increased trend.280differentially expressed genes between rats in model group and normal group (up201, down79) were found by the rat expression microarray analysis, in which genes of amino acid neurotransmitter receptors is24(up14, down10). Compared with the normal group, there is a downward trend of Gabral Gabra4, Gabrb2, Gabrb3, Grin2A expression, and a upward trend of Grm2expression of amino acid neurotransmitter receptors genes in the hippocampus of rats in each group detected by Real-time PCR. The abnormal expression of genes above-mentioned can be rectified after Shu Yu capsule and saikosaponin extract treated.Conclusion:The model rats of depressive disorder with Gan-qi depression can be prepared successfully by using chronicity mild stimulus (CMS),and they can be evaluated effectively by sucrose preference test and open field test. It can be speculated from results of the rat expression microarray analysis that the normal expression of part of amino acid neurotransmitter receptor genes may be the pathogenesis that lead to depressive disorder with Gan-qi depression. The abnormal expression of genes above-mentioned can be rectified after ShuYu capsule and saikosaponin extract treated, and this may be the mechanisms of the effects of Shuyu capsule and Saikosaponin extract.
