Zhuanggu Zhitong Capsule(ZZC) is mainly composed of seven traditional Chinese Medicines, such as Psoralea corylifolia L., Lycium bararum L., Drynaria fortunei(Kunze) J.Sm, Cyathuia officinalis kuan, etc. ZZC is used to tonify liver and kidney, strengthen bone and relieve pain, mainly for the treatment of primary and secondary osteoporosis, and it is effective in clinical. According to the different extraction process, the constituents of ZZC were divided into two fractions. Fraction A included Psoralea corylifolia L. and Ligustrum lucidum Ait., while fraction B contained Epimedium brevicornum Maxim., Drynaria fortunei(Kunze)J. Sm., Lycium bararum L., Cibotium barometz (L.) J. Sm. and Cyathuia officinalis kuan. It was reported that the petroleum ether extract and the EtOH extract exhibited remarkable anti-osteoporosis activity when the researcher screened anti-osteoporosis activity of fraction A of ZZC. The75%EtOH extract was divided into4fractions, which were evaluated for the fractions with anti-osteoporosis activity by ovariectomized rats. In order to identify the components with anti-osteoporosis activity, this subject studied on the petroleum ether extract from fraction A of ZZC, providing a basis for the mechanism research, quality control and second development of ZZC.To separate and purify the chemical constituents of the petroleum ether extract from fraction A of ZZC, several methods had been used in the process including silca-gel column chromatography (CC), recrystalli-zation, sephadex LH-20CC and Pre-TLC, etc. Eight components were obtained from the petroleum ether extract of fraction A of ZZC.Then the anti-osteoporosis activity of the obtained components was investigated by ovariectomized rats. In vivo tests indicated that compound III and IV could significantly increase the bone mineral density and tension of tibia, more strongly than compound V. However, compound II could only increase the tension of tibia, and had a little influence on the bone mineral density of tibia.The structures of compound â…¡,â…¢,â…£ and â…¤ were identified by spectroscopic methods and chemical evidences, which were psoralen, isopsoralen, oleanolic acid and psoralidin. It indicated that psoralen, isopsoralen, oleanolic acid and psoralidin were the active substances of the petroleum ether extract from fraction A of ZZC.In this article, the author identified the active substances of the petroleum ether extract from fraction A of ZZC, and discoveried the anti-osteoporosis activity of psoralidin for the first time.
