Objective: To observe the effect of Zhuanggu Zhitong Capsule on NF-κB signaling pathway of osteoblasts,and to investigate its mechanism in anti-osteoporosis.Method : 60 female 3-month-old SD rats were randomly divided into blank group,model group,Zhuanggu Zhitong Capsule high-dose group,Zhuanggu Zhitong Capsule medium-dose group,Zhuanggu Zhitong Capsule low-dose group and the positive control group,10 rats in each group.Except the blank group,rats in other groups were ovariectomized.Six days after the operation,intragastric administration for one week.Collected blood from abdominal aorta to make serum containing drugs.Osteoblasts from neonatal SD rats were divided into 6 groups after cultured in primary culture,blank group,model group,Zhuanggu Zhitong Capsule high-dose group,Zhuanggu Zhitong Capsule medium-dose group,Zhuanggu Zhitong Capsule low-dose group and the positive control group.Osteoblasts were identified after 3 generations,then intervented by corresponding serum containing drug.The activity of ALP in each group was measured,calcification were observed by alizarin red staining.The protein expression of NF-κB p65 and IκBα were detected by immunohistochemistry and Western blot.Results: Alizarin red staining showed red calcium nodules.The activity of ALP in the model group was decreased compared with the blank group(P<0.01),compared with the model group,the activity of ALP were increased in Zhuanggu Zhitong Capsule low-dose group(P<0.05),Zhuanggu Zhitong Capsule high-dose group and Zhuanggu Zhitong Capsule medium-dose group(P<0.01).Immunohistochemistry and Western blot detected the protein expression of NF-κB p65 and IκBα,the protein expression of NF-κB p65 and IκBα in the model group were increased compared with the blank group(P<0.01),compared with the model group,the protein expression of NF-κB p65 and IκBα were decreased in Zhuanggu Zhitong Capsule high-dose group and Zhuanggu Zhitong Capsule medium-dose group(P<0.01).Conclusion: Zhuanggu Zhitong Capsule can increase the activity of ALP in osteoblasts and decrease the expression of the protein NF-κB p65 and IκBα and play an anti-osteoporosis role.
