Houttuynia cordata have been applied for treatment of respiratory system, alimentary system, urinary system, ophthalmology disease, gynecopathy disease in china for thousands years. However, it faced up many difficulties, because of complex components and mechanism of action.In our study, comparison of Houttuynia cordata volitile oils in root, stem, leaf and flower have done. The volatile oils were obtained with steam distillation. Qualitative and quantitative analysis were done by Gas Chromatography-mass Spectrometry (GC-MS) with areas normalization method.35compounds were identified in root, stem, leaf and flower, among which24components simultaneously exist. The types of the components were similar while the contents were different. The differences and similarities among different parts were analyzed with Principal Component Analysis (PCA), Mahalanobis distance and Correlation Coefficient. The results showed that the difference between the flowers and leaves was little, and the difference between the root and the other three parts was large. The differences of volatile oil from different parts should be fully considered in the use of Houttuynia cordata.GC-MS was used for analysis of volatile oil compounds in serum and tissues, endogenous metabolite in serum before and after oral administration of volatile oil. The change of endogenous metabolite in serum and the distribution of the volatile oil in rat tissue and serum were studied after oral administration of Houttuynia cordata volatile oil. Most of volatile oil compounds were detected in serum and tissue of highest dose groups in vivo. PCA was used to compare endogenous metabolite in the serum of different dosage groups. The PCA classification was significant. The significant difference was shown between the control group and treatment groups.The content of methyl-n-nonylketone at different time in rat after oral administration of Houttuynia cordata volatile oil is detected. The quantity of methyl-n-nonylketone in serum and tissues is determined by GC-MS. There are two absorption peaks at20and40min in serum, two absorption peaks at10and30min in liver, kidney, heart and brain, one absorption peak at20min in spleen, one absorption peak at40min in lung. In the tissue of lung, the content of methyl-n-nonylketone is high and steady.This study provided the foundation of further search about the efficacy, pharmacokinetic and metabolomics studies of Houttuynia cordata.
