Objective In order to explore the inner relationship between the protein polymorphism and variation of volatile constituents in Houttuynia cordata. Protein polymorphism and Volatile Constituents of fresh Houttuynia individuals which were chosen from Good Agricultural Practice(GAP) condition were studied in this research.Method The total proteins extracted from fresh Houttuynia individuals in the same GAP were detected by using SDS-PAGE gel electrophoresis technology, dual wavelength TLC scanner were used to depth scan on the gel electrophoresis of protein bands. With peak area as indicators, total quantum statistic moment theory were used to statistical analysis on protein fingerprinting, and section partition with standard protein Marker as control. At the same time, volatile oil of each fresh Houttuynia individual were determined using GC-MS after extraction of total protein. The common components of volatile oil in each Houttuynia cordata were associated with polymorphic protein sections.Result (1) The analysis results of using the thin-layer scanner scan gel protein electrophoresis, The RSD of AUCT, MCRTT and VCRTT is40.92%,6.01%and18.57%, respectively. These show that there are great differences of total protein between different strains of Houttuynia cordata.(2)The GC-MS results of volatile constituents shows that41-75compounds were detected from volatile oil between different strains of Houttuynia cordata, and the relative content of70kinds of volatile constituents were more than0.05%, including23common components of volatile oil, and the range of variable coefficient of the relative contents is between13.28to53.99. Those suggest that there are great differences both classifies and relative contents between the volatile constituents of different strains of Houttuynia cordata when they have the same growth environment, collecting time and processing method.(3) The results of correlation between the common components of volatile oil and each section of its protein show that the different protein segments affect on different component contents of volatile oil and the same protein segment for the component contents of volatile oil are also vary. Therefore, the differences of the chemical component on the species and quantity were directly influenced by protein polymorphism.Conclusion There are rich diversity between protein and volatile constituents of different strains of Houttuynia cordata in the same GAP, and there are the inner relationship between the protein polymorphism and variation of volatile constituents, Which lay a foundation for looking for the key (proteins) enzyme that can control the volatile constituents of Houttuynia cordata. Therefore, we can’t ignore the effect of protease in the quality control of Chinese medicine.
