objective:To understand the application of rifapentine capsulecan increase or inhibit the immune function of remove tuberculosis, andwill preliminary study the possible mechanism of the changes onimmune function. Methods:1.Take70male SD rats, weighing180±30g,were randomly divided into four groups, respectively, before feeding,feeding10only, feeding a month, feeding three months, feeding threemonths after drug withdrawal a month, after the three groups were20,experimental group(10only), control group (10only)2. before Feedingwith free feeding, without any intervention, the other three groups: theexperimental group with rifapentine capsule gavage, the control groupwith normal saline lavage, more than the same feeding conditions.3.Using flow cytometry detection of CD3+, CD4+, CD8+and NK cell counts,using carbon particle clearance assay for the detection of phagocyticfunction of macrophages Results:1.In the Comparison of parameters ofexperimental group and the control group. Comparison of parameters infeeding one month and feeding three month after drug withdrawal amonth, the index showed no significant differences. In the feeding threemonth later, the Comparison of parameters in the experimental groupand the control group, the differences in CD3+、 CD8+、clearance index,average phagocytosis rate were statistically significant between the two groups; the CD4+, CD161+, CD4+/CD8+had no significant difference2.The control group at all time points compared with the blank controlgroup, except CD3, average rate, phagocytic clearance,The difference ofother index in feeding one month, three month, feeding three monthlater after drug withdrawal a month and blank group has statisticssignificance,3. experimental groups at each time point compared with theblank control group: the difference of CD3+, CD8+, CD4+/CD8+infeeding one month, three month, feeding three month later after drugwithdrawal a month and blank group has statistics significance; CD4+,CD161+, average rate, phagocytic clearance index at each time point hasno significant difference; Conclusion:1.in this experiment, theexperimental group and the control group comparison, feeding one monthand feeding three month later after feeding three month later after drugwithdrawal a month, the index showed no significant differences,indicating that rifapentine capsule efficacy does not reach the effect onthe immune function of strength on the feeding one month group, feedingthree month later after drug withdrawal a month may be due to stopfeeding, after stopping the medication in vivo reduction or afterwithdrawal the original drug varies with the in vivo metabolic clearance,2feeding three month later,the experimental group and the controlgroup, The differences of CD3+, CD8+,clearance index, averagephagocytosis rate were statistically significant, In the experimental group, the mean standard deviation rate of CD3+, CD8+, clearance index,average phagocytosis was significantly higher than that of control groupincreased, and each index had a significant difference, indicates that withthe feeding time, drug for individual immunity indices have certain effect.3. in the control group at all time points compared with the blank controlgroup, except CD3+、 average rate,、phagocytic clearance index,Thedifference of CD4+index、CD4+/CD8+、CD161+in the feeding one month、feeding three month、 feeding three month later after drug withdrawal amonth and blank group were statistically significant, the difference maybe due to sampling error or errors caused by experimental operation;4.experimental groups at each time point compared with the blank controlgroup The difference of CD3+、 CD8+、CD4+/CD8+in the feeding onemonth、feeding three month、 feeding three month later after drugwithdrawal a month and blank group were statistically significant,indicating that the effection of drug on related indexes was positivelyrelated to the acting time of the drug. CD4+, CD161+, average rate,phagocytic clearance index at each time point has no significantdifference, can not rule out the drug had no effect on these indexes.
