ObjectiveTo investigate the immunomodulatory activity of resveratrol(RES),spleen lymphocytes and peritoneal macrophage from the mice were taken as target cells in vitro.The immune function model was established for studying the immunomodulatory activity of resveratrol from cellular and molecular levels.And these would provide experimental basis for further development and utilization of resveratrol.Methodhe immune function model was established for studying the immunomodulatory activity of resveratrol used to spleen lymphocytes and peritoneal macrophage from mice in vitro.The effect of cell proliferation was detected by MTT method in the different concentration RES groups,RES +concanavalin A(Con A)group and RES+Lipopolysaccharides(LPS)group.The effect of spleen lymphocytes secrete cytokines was detected by ELISA kits in the different concentration RES groups.The effect of expression level of cytokine gene was detected by RT-PCR method in the different concentration RES groups to spleen lymphocytes.ResultStimulation index(SI)of the spleen lymphocyte was increased in the different concentration RES groups,RES+Con A group and RES+LPS group.And SI of RES+Con A group and RES+LPS group was higher than that of RES groups.RES could stimulate splenic lymphocytes and increase cytokinessecretion of IL-3,IL-4,IL-12 and IFN-?,and up-regulate the cytokines gene level of IL-3,IL-4,IL-12 and IFN-? in spleen.The activity of the peritoneal macrophages was detected by devouring staphylococcus aureus.The function of macrophage devouring staphylococcus aureus was increased in the different concentration RES groups,RES+Con A group and RES+LPS group.And the effect of RES+Con A group was higher than that of the different concentration RES groups.RES+LPS group was lower than that of the different concentration RES groups.Conclusion1?Resveratrol could single stimulate the proliferation of spleen lymphocyte and conjunction with Con A or LPS.2?Resveratrol could increase the secretion of cytokines IL-3,IL-4,IL-12 and IFN-? by increasing the m RNA gene expression of IL-3,IL-4,IL-12 and IFN-?,so as to enhance the immune function of lymphocytes.3?Resveratrol could enhance the phagocytic function of peritoneal macrophages.
