The methylenetetrahydrofolate reductase (methylenetetrahydrofolate re ductase, MTHFR) can catalyze5,10-methylenetetrahydrofolate into5-me thylenetetrahydrofolate. MTHFR is a key enzyme of folic acid and meth ionine. In the metabolism of homocysteine, it plays a key role. MTHFR genes have polymorphism, and its mutations will make MTHFR thermo stability and enzyme activity declining. At last it will cause high homoc ysteine hematic disease. It will affect DNA synthesis and methylation. A lot of studies have a demonstration that high homocysteine is a risk factor of stoke.Objective1. The correlation between methylenetetrahydrofolate reductase gene polymorphism (MTHFR) and stroke of hubei people was discussed.2. The correlation between methylenetetrahydrofolate reductase gene polymorphism (MTHFR) and stroke of TCM syndrome type was discus sed.Methods70patients of stoke were selected, and were divided into4groups a ccording to the syndrome differentiation-typing TCM:Gan yang shang k ang syndrome group, Tan zhuo yong zhi syndrome group, Qi xu xue y u syndrome group, Yin xu feng dong syndrome group. The patients of stoke group were divided into the sthenia syndrome group and the asthe nia syndrome group, according to the syndrome differentiation-typing T CM, all the research objects are phlebotomize and extracting DNA to Polymerase Chain Reaction-united restriction Fragment Length Polymorp hism method (Polymerase Chain Reaction-restriction-Fragment Length Po lymorphism, PCR-RFLP) test methylenetetrahydrofolate reductase gene C677T, gene Polymorphism A1298C sites, to compare the patients (group S) and normal control group (group C); the sthenia syndrome group an d the asthenia syndrome group; TCM syndrome differentiation between each card type of methylenetetrahydrofolate reductase C677T and A1298C genotype distribution frequency and the distribution of allelic frequen cy.Results1. Significant differences were observed in the frequencies of C677T genotypes between the S and C groups. The C677T genotype frequen cy and the T allelic frequency of C677T in S patients were higher than that of the normal controls.(P<0.05)2. Significant differences were observed in the frequeneies of A1298C genotypes between the S and C groups. The A1298C genotypesfrequ ency and the C allelic frequency of A1298C in S group were higher th an that of the normal controls.(P<0.05)3. The patients of S group were divided into the sthenia syndrome group and the asthenia syndrome group according to the Syndrome Dif ferentiation-typing TCM. The frequeneies of C677T and A1298C genoty pes are all consistent with Hardy-Weinberg law.(P>0.05)4. No significant differences were observed in the genotype and all elic frequencies of MTHFR gene C677T between the sthenia syndrome group and the asthenia syndrome group.(P>0.05)5. No significant differences were observed in the genotype and all elic frequencies of MTHFR gene A1298C between the sthenia syndrome group and the asthenia syndrome group.(P>0.05)6. The patients of stoke group were divided into four groups accor ding to the syndrome differentiation-typing TCM. No significant differen ces were observed in the genotype and allelic frequencies of MTHFR g ene C677T and A1298C among the four syndrome groups.(P>0.05)Conclusion1. The C677T genotypes were associated with the presence of stok e.The T allele of C677T is a geno-factor while the T allele is the harm ful factor.2. The A1298C genotypes were associated with the presence of sto ke.The C allele of A1298C is a geno-factor while the C allele is the harmful factor.3. The C677T in MTHFR gene were not associated with the syndr ome differentiation-typing TCM of stoke and the sthenia syndrome grou p and the asthenia syndrome group of stoke.4. The A1298C in MTHFR gene were not associated with the synd rome differentiation-typing TCM of stoke and the sthenia syndrome grou p and the asthenia syndrome group of stoke.5. The C677T and A1298C in MTHFR gene were not associated w ith the syndrome Differentiation-typing TCM of stoke.
