| Objective: To observe the effect of three Xinjiang plants (fenugreek, chickpeas andpomegranate rind) water extract on peptide peptidase IV (DPP Ⅳ) activity in vitro andin vivo experiments. Methods:1.Experiment is using DPPâ…£inhibition model, the threeXinjiang a hypoglycemic effect of plant extracts were screened in vitro.2.Selected90male mice, injection of streptozotocin (STZ)180㎎·ãŽ-1in traperitoneal, establishmentof a diabetic mouse model in vitro, the model mice were randomly divided into modelgroup, treatment group, including the sitagliptin group, fenugreek low dose group (3g·ãŽ-1), calabash Ba middle dose group (6g·ãŽ-1), fenugreek high dose group (12g·ãŽ-1),chickpea low dose group (1g·ãŽ-1), chickpea middle dose group (2g·ãŽ-1), chickpeas inhigh dose group (4g·ãŽ-1), determination of mouse portal blood serum GLP-1content andDPPâ…£ activity effect, and the determination of related biochemical parameter aftergiving4weeks. Results: Fenugreek extract water IC500.03mg·mL-1, extraction frompomegranate peel IC500.19mg·mL-1, chickpea extract IC500.09mg·mL-1.Compared whitthe normal control group with model group mice, the model group of weight significantreduces, fasting plasma glucose, total cholesterol, triglyceride, low density lipoproteinincreases, high density lipoprotein, peptide C levels decrease, decline in insulin secretion,insulin resistance increased, GLP-1 decreased(P<0.05,P<0.01). Fenugreek and chickpeawas administered for4weeks, compared with model group, fasting plasma glucose,totalcholesterol, triglyceride, low density lipoprotein decrease, high density lipoproteinsincreased significantly, C peptide levels increased, increased insulin secretion, insulinresistance decreased, with the rate of DPP IV inhibition increased, the content of GLP-1was increased(P<0.05,P<0.01). Conclusions: Experiments decreased the blood glucoseand blood lipid in diabetic mice, the possible mechanisms of action were dipeptidlypeptidase IV inhibitory activity in Xinjiang sugar aqueous extracts of plants and so on. |
