Qiweibaizhu Powder protective effect on HRV infection of suckling mouse liver tissueObjective:Explore HRV infection of suckling mouse hepatic tissue pathological and its functional changes to clarify the mechanism of protection of traditional Chinese medicine Qiweibaizhu Powder on the liver tissue of suckling mice infected.Method:1. The plaque formation test to determine virus titer. Using plaque forming unit said virus titer. HRV suspension infected with5-day-old suckling mice by the oral route. Taking the suckling mice feces by monoclonal antibody enzyme-linked immunosorbent assay (McELISA) confirmed the rotavirus antigen-positive, Established neonatal rat infecting HRV model.2. suckling mice were randomly divided from the same litters into model group (MG), Qiweibaizhu Powder group (BG), ribavirin group (ZG), and established the normal control group (NG), were the total of four groups. Neonatal rat of each treatment group were treated with the drug by oral drop50μl/(only time),3times/day. Normal control group and model group drops the same amount of cells to maintain the liquid by oral.3.(1)5days treatment of suckling mice decapitated and taken blood. The blood was sended to test liver function, suckling mice liver tissue did pathological section,HE staining and electron microscopy, observed the changes in liver pathology, liver cell ultrastructure.(2) Determined the content of IL-2, IL-8and TNF-a and IFN-yin the drug intervention suckling mice serum by ELISA. Using RT-PCR method to determine IL-2, IL-8, TNF-alpha, IFN-y mRNA in suckling mice liver tissue. Results:1. suckling mice infected with HRV for5days, The differences of AST, ALT content in BG suckling mice serum and ZG had not statistically significant(P>0.05), But the differences of AST, ALT content in BG suckling mice serum and MG suckling mice serum had statistical significance(P<0.05).2. The structure of normal suckling mice liver tissue clear and has not inflammatory cell infiltration under ordinary biological optical microscope; suckling mice liver tissue congestion and liver cell degeneration, has inflammatory cell infiltration in model group; Suckling mice liver tissue has mild hyperemia and vacuolar degeneration of liver cells in ribavirin treatment group; Suckling mice liver tissue have also mild hyperemia in Qiweibaizhu Powder group, but inflammatory cells are rare. Morphology of normal neonatal rat liver cells is normal in electron microscope, and cristae clear, has glycogen granules in the cytoplasm, complete mitochondria can be seen. Suckling mice liver cells has lipid particle deposition in the Model group, Bile pigment appears, Liver cells have become pyknotic, cracking, microvilli fall off, mitochondria obviously swell, has visible empty bubble formed, Liver capillary bile duct expansion, cholestasis has visible. Part of the rough endoplasmic reticulum swellen in ribavirin group of neonatal rat liver cells, Mitochondria has vacuole and swolling, Liver cells focal dissolved and has vacuolization. The neonatal rat liver cells has focal dissolved in Qiweibaizhu Powder group, has a small amount of vacuolar degeneration, the rough endoplasmic reticulum dilatation in liver cells, mitochondria Swelling is visible.3. Compared of IL-2, IL-8and TNF-a and IFN-γ expression in suckling mice serum:IL-8, TNF-a content in Qiweibaizhu Power was significantly lower than the model group(p><0.05), There is no significant difference compared with Ribavirin group(P>0.05),There is significant difference compared with normal group(P<0.05),The content higher than the normal group;IL-2, TNF-y content in Qiweibaizhu Power was significantly higher than the model group(P<0.05), There is no significant difference compared with Ribavirin group(P>0.05), There is significant difference compared with normal group(P<0.05), The content higher than the normal group;IL-8mRNA, TNF-a mRNA in IL-2mRNA and IFN-y mRNA expression in neonatal rat liver tissue and ELISA results are similar.Conclusion:1. Qiweibaizhu Powder can reduce AST, ALT and TNF-a content in HRV infection of suckling mice serum, and reduce neonatal rat liver tissue lesions.2. Qiweibaizhu Powder can promote IL-2, IFN-y expression in HRV infection of suckling mice liver tissue, clear the infection the HRV.
