ObjectiveOptimizing and determining the extraction process of eucommia key-fruit seed oil by supercritical fluid extraction. Optimizing and determining the extraction process of aucubin in eucommia key-fruit seed by CO2supercritical fluid extraction with entrainers. Investing and determining the optimal processing parameters for eucommia pure rubber extraction from eucommia key-fruit.MethodsBased on single factor preliminary experiment, the experiments parameters were selected. According to the principles of Box-Benhnken experiment design, three factors and three levels response surface methodology was used. According to the regressive analysis, the influencing factors of the processes were determined. By using the extracting rate of eucommia key-fruit seed oil as response value, the response surface and contour were drew. After analyzing the significant and interaction of each parameter, the optimal extracting processing conditions were determined, of eucommia key-fruit seed oil.Based on single factor preliminary experiment, the experiments parameters were selected. According to the principles of Box-Benhnken experiment design, four factors and three levels response surface methodology was used. The effects to extracting ratio of aucubin were analyzed, from four parameters of extracting pressure, extracting temperature, extracting time and content of Ethanol entrainer, and the optimal extracting processing conditions were gotten for aucubin extracting from eucommia key-fruit seeds meal.The non-rubber components of eucommia key-fruit nut shell were decomposed by alkali extracting. By extracting eucommia rubber with petroleum ether, and using gel-freezing method to precipitate the eucommia rubber, the optimal processing parameters were determined for eucommia pure rubber extraction.ResultsThe optimal extracting processing condition for eucommia key-fruit seeds oil were as followed:extracting pressure33MPa, extracting temperature40℃, extracting time54min, and the extracting ratio could reach26.64%.The optimal extracting processing condition for aucubin in eucommia key-fruit seeds meal were as followed:extracting pressure27MPa, extracting temperature50℃, extracting time120min, and the extracting ratio could reach26.64%, and content of Ethanol entrainer80%, and the average extracting ratio could reach1.9510%(RSD=0.696%) Used the alkali extracting method to treat the eucommia key-fruit nut shell with10%alkali solvent in90℃for3h, and used Soxhlet extraction method to extract with petroleum ether in85℃for27h, and then the extract was frozen in0℃for1h to precipitate the eucommia rubber. The extracting rate of eucommia rubber in eucommia key-fruit nut shell is20.48%, and5.47%in eucommia key-fruit, and the purity of extracted eucommia rubber is87.52%.ConclusionBy using response surface methodology, studied the parameters optimization and supercritical fluid extraction conditions regression of eucommia key-fruit seeds oil. The result, by variance analysis, showed that the model was significant (p<0.0001), adjusted determination coefficient0.9315, and could predict the rules of extracting ratio of eucommia key-fruit seeds oil changed with the parameters, and establish the theoretical basis and technical basis for further development and utilization of eucommia key-fruit resources.By using response surface methodology, studied the parameters optimization and supercritical fluid extraction conditions regression of aucubin in eucommia key-fruit seeds meal. The result, by variance analysis, showed that the model was significant (p<0.0001), adjusted determination coefficient0.9915, and could predict the rules of extracting ratio of aucubin changed with the parameters. The study showed that supercritical fluid extraction method for aucubin extraction was simple and convenient with good yield, and was a high extracting method for aucubin extraction. The study established theoretical basis for aucubin industry extraction of eucommia key-fruit seeds meal and has practical guiding significance.Eucommia key-fruit has high content of eucommia rubber, and by using alkali extraction method for decomposing, extracting with petroleum ether is feasible. By using gel-freezing method to precipitate the eucommia rubber, with light yellow, the purity of the rubber reaches87.52%. To study the eucommia rubber extracting process in eucommia key-fruit, can establish the experimental basis for eucommia rubber in eucommia key-fruit resources further development and utilization.
