Research Of The Effects And Action Mechanism Of Eucommia Ulmoides Oliv.Seeds Draff On The Postmenopausal Osteoporosis

Eucommia ulmoides Oliv is a traditional kidney-tonifying herbal medicine with a long safe use history for treatment of bone fractures and joint diseases in China. Ordinarily Eucommia ulmoides Oliv bark and leaves was used for osteoporosis. According to the historical Chinese herb-medicine book,bencaotujing, Eucommia ulmoides Oliv seeds was also appropriate as medical material. We intend to optimizie the extractive condition of Eucommia ulmoides Seeds to separate and purify Eucommia ulmoides Seed’s Extract. And the influence of ESE on bone formation would be investigated by cytological experiments. On the overall level, the effects and action mechanism of Eucommia ulmoides Oliv.seeds draff on the postmenopausal osteoporosis will be inspected.By means of combining ultrasonic wave-assisted extraction and maceration, we have optimized the extraction technology of Eucommia ulmoides seeds. Meanwhile, through the method of MTT, the activity measurement method of ALP and Alizarin red staining, it can be evaluated about the effects of ESE on proliferation, differentiation and mineralization of osteoblasts.Bilateral ovaries removing surgery was used to built OVX rat model.The bone de nsity of OVX rats was measured by dual-energy X-ray absorptiometry. The three point bending test were used to determinate the crush force, femur fracture strength and other indicaters.The microstructure of femoral bone trabecular observed by Micro-CT willbe analy zed. Enzyme-linked immunosorbent assay was used for measuring serum estrogen level and biochemical markers of bone metabolism. Automated biochemical analyzer was used to measure the contents of calcium, phosphorus, creatinine and ALP in blood and urine. Through all the test results, we are aiming at evaluate the influence of ESE on OVX rats.After optimization, the extraction yield of aucubin has achieved69.19%.Aucubin is the active component of ESE to promote the proliferation, differentiation and mineralization of osteoblasts, and the best concentration is25.6ng/ml. Beyond25.6ng/ml, the effects will weaken. According to the research, BMD of ESE medication administration team is obviously higher than OVX team. And Micro-CT, experiments of bio-mechanical property, and the biochemistry analyses results shows the crush force, femur fracture strength, Tb.N., Tb.Sp., Tb.Th. and the level of E2, ALP, OC, OPG, RANKL in blood of medication administration team is higher than OVX team, but the level of U-Ca, U-P, U-Cr, U-Ca/U-Cr, U-P/U-Cr in urine is lower. We can conclude that the extraction of Eucommia ulmoides seeds can promote the proliferation and differentiation of osteoblasts, restrain bone resorption, increase the BMD of osteoporotic modle rats. This provides the basis on the development of Eucommia ulmoides seed to therapy postmenopausal osteoporosis.
