ObjectiveObserved the inflammatory factors in the change on vascular dementia rats,anddiscuss the cerebral protection of the berberine on vascular dementia rats.MethodsTwo vessel occlusion using the modified method of Vascular Dementia rat modelprepared by sodium nitroprusside,36male sprague dawley rats were randomly dividedinto six groups: normal group, sham operation group, dementia model group (modelgroup), low dose of berberine group(10mg/kg) treatment group,middle dose ofberberine group(20mg/kg) treatment group,high dose of berberine(40mg/kg) treatmentgroup. After continuous administration for4weeks with different drugs.Though theMorris water maze detecting the rats’ behavior and determination of the hippocampus inthe IL-8, IL-1β, TNF-α levels.Results1. Compared with the normal group, sham group no obvious behavioral disorders,model group and treatment group had significant barriers to learning and memorybehavior.The hippocampus IL-8, IL-1β, TNF-α expression of model group andtreatment group than the normal group, sham operation group was significantly higher(P <0.05).2. Compared with model group, treatment group compared with the model ofbehavioral symptoms was significantly improved.In the treatment group hippocampusIL-8, IL-1β, TNF-α expression was significantly decreased (P <0.05).More decreased inthe high dose group than the middle and the low treatment group was significantly (P<0.05).ConclusionInflammatory response involved in the occurrence of vascular dementia; berberinecan improve the behavioral symptoms of rats with vascular dementia, berberine thatcan play a role in brain’s protection may be inhibit the inflammatory response pathwayby reducing the inflammatory factors (such as IL-8, IL-1β, TNF-α, and so on), so that the symptoms of vascular dementia improved effectively. Berberine’s inhibition ofinflammatory response and brain protection in a dose dependent phenomenon.
