Vascular dementia, vascular dementia (VD) refers to ischemic and hemorrhagic cardiovascular and cerebrovascular diseases and brain dysfunction caused by disease, which produces acquired intelligent damage, especially with memory loss syndrome, senior cognitive abilities give priority to, as a chronic progressive disease. In recent years, more and more attention about immune inflammatory lesions hypothesis and free radical damage hypothesis in pathological mechanism in the role. That's more than a brain disease, kidney disease, and which is the relation of the disease, the qi deficiency of Yin and Yang, qi and blood, to reality, sputum marking to wet miscellaneous, blood stasis stranded. This research is to drug Duhuo for the starting point, use on nimodipine is discussed in vascular dementia (VD) immune inflammatory lesions and free radical damage.Purpose:This study aims to explore drug Duhuo in the intervention vascular dementia rat models of immune inflammatory lesions and oxidative damage, main observation model rat tumor necrosis factor alpha (TNF alpha), superoxide dismutase (SOD) and malondialdehyde (MDA), this paper discusses drug Duhuo delay mechanism of the influence of the aging brain, the mechanism for the brain aging research and treatment of vascular dementia medicine to provide scientific experiment basis.Materials and methods:Adopt double side carotid artery ligation of lasting method (2 - VO) shaping chronic cerebral perfusion shortage of vascular dementia model. Giving Duhuo, western medicine on nimodipine, physiological saline lavage intervention. Four weeks later, using the water maze experiment to observe learning and memory ability of vascular dementia models rats. And by the morphological changes of model rats hippocampal CA1, using immunohistochemistry methods to detect expression of TNF alpha of rat hippocampal CA1 area. Using enzyme-linked immunosorbent double antibody clip method (ELISA), to detect the differences of each rat serum TNF alpha, MDA and SOD's expression.Results:1. In the water maze experiments, traditional chinese medicine group and western medicine group after ginving drug compared with before, the escape times were significant shorter (P<0.01), and Duhuo and nimodipine on bilateral carotid artery ligation VD model of rats have increased level of learning and memory.2. TNF alpha ELISA test results: compare with blank control and sham operation group, the expressions of TNF alpha of the model group, western medicine group and traditional chinese medicine group are significantly went up(P<0.01). Compared with the model group, the traditional chinese medicine group and western medicine group TNF alpha express all declined (P<0.05). Between blank control and sham operation group, between traditional chinese medicine group and western medicine group, there are not any differences in statistics.3. TNF alpha immunohistochemical testing results: compare with blank control and sham operation group, the model group, western medicine group and the traditional chinese medicine group have the positive cells expressed obviously improved (P<0.01). Compared with the model group, the express of positive cells of traditional chinese medicine group and the western medicine group have decreased obviously (P<0.01). Between blank control and sham operation group, between traditional chinese medicine group and western medicine group, there are not any differences in statistics.4. SOD, MDA test results: compare with blank control and sham operation group, the model group, western medicine group and the traditional chinese medicine group have obvious drop expression of SOD (P<0.01), the expression of MDA rose significantly (P<0.01). Compared with the model group, traditional Chinese medicine group and the western medicine group's expression of SOD were increased , (traditional chinese medicine group P<0.01),( western medicine group group P<0.05). Chinese traditional medicine group and the western medicine group's MDA expression all declined,( western medicine group P< 0.01), (chinese traditional medicine group P<0.05). Between blank control and sham operation group, between traditional chinese medicine group and western medicine group, there are not any differences in statistics.Conclusions:1. Adopt double side carotid artery ligation of lasting method shaping chronic cerebral perfusion shortage of vascular dementia model of rats, the escape times were significant longer, and reduced levels of learning and memory. Rats serum TNF alpha, MDA's expression heighten and SOD's expression descend, rats'hippocampal CA1 area TNF alpha positive cells express increase, these might be vascular dementia model of pathological changes of rats.2. Duhuo can improve the learning and memory level of model rats, restrain rats serum TNF alpha,MDA's expression and improve SOD's expression , restrain rats'hippocampal CA1 area TNF alpha expression, Duhuo can suppress the immune inflammatory lesions and free radical damage of VD model rats, it may be a mechanism of Duhuo for prevention of vascular dementia.
