| Objective To examine the changes of renal tissue structure and the expression of hepatocyte growth factor(HGF)and hepatocyte growth factor receptor (c-met) in renal tissue of neonatal rats exposed to prolonged hyperoxia and to observe the influence of astragalus injection on the renal damage.Methods With exposure to prolonged hyperoxia induced lung injury in neonatal rat model. Neonatal Sprague-Dawley (SD)rats (n=144)were randomly divided into4groups within12h after birth. Group â… (n=36):room air+normal saline(NS); group â…¡ (n=36):room air+astragalus injection; groupâ…¢(n=36):hyperoxia+NS; groupâ…£(n=36): hyperoxia+astragalus injection.Groups â… and â…¡ were placed in room air; gorupsâ…¢andlVwere exposed to hyperoxia.Groups â… andâ…¢of intraperitoneal injection of NS, groups â…¡ andâ…£of intraperitoneal injection of astragalus injection(10g/kg). On day3th,7th andl4th day after birth,8rats in each group were killed. The changes of renal tissue structure were examined by hematoxylin-eosin(HE) staining method; the protein expression levels of HGF in the renal tissue was detected by immunohistochemical method; the mRNA expression intensities of HGF and its receptor(c-met) were measured by reverse transcription polymerase chain reaction (RT-PCR).Resultsâ‘ Compared with groups â… , â…¡ andâ…£, groupâ…¢of visible mesangial cell proliferation, renal tubular epithelial cells swelling, inflammatory cells infiltration, with the passage of time, the pathological changes more and more obvious. The pathological changes of groupâ…£ were more significantly reduced than group â…¢.(DCompared with group â… and group â…¡, the protein expression levels of HGF were enhanced after3days in groupâ…¢(P<0.05),7d were significantly enhanced(P<0.05),14d were enhanced too(P<0.05), but at14d, the enhanced tendency were weaker than at7d; Compared with group â…¢, group â…£ of all time points were significantly enhanced(P<0.05); No significantly difference between group â… and group â…¡ at all time points(P>0.05).â‘¢HGF and its receptor (c-met) mRNA expression intensities and theirs protein expression levels were similar to the changing tendency. HGF receptor(c-met) mRNA expression intensities were higher than HGF in all groups at the same time points. Conclusion Astragalus injection can promote expression of HGF and its receptor(c-met) in hyperoxic injuried renal tissue and improve renal tissue injury and promote the impaired renal tissue recovery. |
