Objective To study the correlation between the single nucleotide polymorphism of rs9939609in FTO (Fat mass and obesity-related gene) and coronary heart disease (CHD) patients with type2diabetes mellitus (T2DM).Subjects and Methods Random selection of Endocrinology, Affiliated Hospital of Jining Medical College in July2010to March2011,518cases of type2diabetic patients hospitalized by the Department of Cardiology,260male cases and258female cases, the average age(58.07±9.44); divided into2groups according to whether the merger of coronary heart disease, in which type2diabetes mellirus (T2DM group) of256cases,124male cases and132female cases, the average age(57.34±9.63) years with type2diabetes262cases of coronary heart disease group (T2DM+CHD group),136male cases,126female cases, the average age(58.80±9.25), coronary heart disease duration of less than type2diabetic duration. Type2diabetes was diagnosed1999WHO criteria, to exclude type1diabetes and secondary diabetes; coronary heart disease diagnosis is also according to the WHO diagnostic criteria. All subjects were required liver, with normal renal function, to exclude thyroid disease, blood disease, peripheral vascular disease, and under examination at least one month can not take lipid lowering drugs. Control group (NC group) healthy Affiliated Hospital of Jining Medical College242cases,128male cases,114female cases, the average (57.27±10.64) years to rule out diabetes, coronary heart disease, high blood pressure, high cholesterol, kidney, etc. All of the subjects were in Shandong Province Han, unrelated, body mass index (BMI) above18.5kg/m2was associated. Conducted a questionnaire survey of all subjects, including age, marital status, income, nutritional status, lifestyle, and measurement of height, weight, blood pressure (BP), waist circumference (WC). In accordance with the BMI All subjects were divided into normal control group (18.5kg/m2≤BMI <25kg/m2), overweight (25kg/m2≤BMI<28kg/m2), obesity (BMI≥28kg/m2). Specimens from fasting blood samples for genomic DNA was extracted and biochemical characteristics. Using the automatic biochemical analyzer (Sapphire600) Determination of blood lipids, blood sugar, blood lipids, including triglycerides (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL) and low density lipoprotein cholesterol (LDL). Application of TaqMan-MGB probe allele-specific hybridization analysis detected the FTO rs9939609single nucleotide polymorphisms, to calculate the allele frequency, clinical indicators of differences in analysis of the FTO gene polymorphism with type2diabetes and type2diabetes with coronary artery disease.Results1General information T2DM group with T2DM+CHD group of patients with BMI, WC, systolic blood pressure (SBP), diastolic blood pressure (DBP), HbAlC FBG and FINS and HOMA-IR,2hBG, TG and the TC and LDL were significantly higher than that of NC group, while HDL was significantly lower than the NC group (P<0.05); of T2DM+CHD group, the SBP, HbAlC,2hBG, TC and LDL were higher than that of T2DM group, the HDL was less than the that of T2DM group (P<0.05).2The performance of the population of the FTO rs9939609genotype TT, TA, AA. Three genotypes distribution of frequency in the population were75.92%,22.37%,1.71%. A and T allele frequency distribution in the population of12.89%and87.11%. A allele and the TA+AA genotype in the T2DM group and T2DM+CHD group were significantly higher than the that of NC group (P<0.05), grouped by BMI, the obese group A allele frequency was significantly higher than that of the normal control group (P<0.05). 3Genotype on the clinical indicated that people with TA/AA genotype, BMI, and WC, the TC and LDL, WC, FINS and HOMA-IR, SBP,2hBG level were significantly higher than people with TT genotype, the HDL was significantly lower than that with TT genotype (P<0.05).4Type2diabetes, type2diabetes with coronary heart disease as the dependent variable, the T allele, A allele, AA, TA, and TT genotypes for the argument step by step logistic regression analysis results showed that A allele was related with type2diabetes and type2diabetes with coronary heart disease.Conclusions1FTO rs9939609locus A allele was associated with type2diabetes and type2diabetes with coronary heart disease related to the occurrence, indicating that the A allele may be risk allele of Jining Han population with type2diabetes and type2diabetic patients with coronary heart disease.2The results of this study showed the obese group the FTO rs9939609A allele frequency was significantly higher than the normal control group, confirmed the FTO rs9939609A allele was related with BMI.3Type2diabetes and type2diabetes with coronary heart disease was not only the existence of glucose and lipid metabolism disorders, but also there was a certain degree of insulin resistance.
