Object To establish a reliable and stable model of dilated cardiomyopathy-heart failureof rats by injecting adriamycin into their caudal veins. Then we examine the strictionand diastole function of their hearts with high frequency echocardiography, and evaluateits value in the research of dilated cardiomyopathy-heart failure of rats caused byadriamycin.Methods the way to establish the model:30male, healthy SD rats were dividedrandomly into two groups. The ADM group was made into the model of heart failure byinjecting ADM into their caudal veins (2mg/kg body weight, once a week, for6weeks),while the normal control group was made by injecting the same amount of physiologicalsaline. The bite and sup, the mental state, the change of the fur, the change of abdominalcircumference and body weight of the rats were observed every day during themedication process. Two weeks after the last medication, the structure of heart of twogroups were observed with high frequency echocardiography, the indexes of heartfunction were measured and then the images were stored and recorded. The rats wereexecuted after the examination. We got their hearts, and then got rid of the tissue ofatriums, right ventricles and great vessels. The left ventricles were fixed with formalinfirst, and then embedded with paraffin. At last the muscle of left ventricles was stainedin the HE staining method and the VG staining method.By using Image-Pro plus6.0which was a software of quantitative analysis to analyze two groups of the VG stained sections, two groups of collagen volume fraction (CVF)of cardiac muscle were gotten. Next, we analyzed the simple linear relation betweenCVF and EF and FS of rat cardiac.Results The ADM group of rats which were still alive after8weeks presented anobvious symptom of heart failure: there was a decrease in the activity and diet, a badmental state, dropped fur in different degree, increased abdominal circumference andwhen their position was changed, the movement of the liquid in their abdominal cavitycould be observed, and their resistance decreased when were caught. On the contrast,the activity, the mental state, the fur and the diet of the normal control group changedlittle. When they were executed, we found the volume of hearts of the ADM group wasbigger than that of the normal control group, and there was varying degree of hematodeshydrothorax and hydroperitoneum of the ADM group, but not of the normal controlgroup.Compared with the circumstances of the normal control group, the striction and diastolefunction of the hearts of the ADM group obviously descended: IVS and LVPW becamethin, LV inner diameter enlarged, EF and FS decreased, and there was a statisticssignificance between the difference(P<0.01). The frequency spectrum of Mitral Valveshowed that Peak E/Peak A<1.Compared with the normal control group, the HE staining of the muscle of leftventricles of the ADM group presented that the cells of myocardium ranged disorderly,and the cells were amplified, having hydroncus. The VG staining presented that theintercellular substance of cardiac muscle of rats was fibrillated obviously. However, thevolume and the shape of the muscle cells of left ventricles of the normal control groupwere regular and the cells were clear and in good order, while the fibril content ofintercellular substance of cardiac muscle was in a normal range.By analyzing the VG stained sections of left ventricles with Image-Pro Plus6.0, two groups of CVF would be gotten. The CVF of ADM rats was greater than the normalcontrol group, while having a statistics significance(P<0.01). There was a nicenegative linear correlation between CVF and EF, FS.Conclusions1.The change of behavior of rats, the general sample, the emerging ofhydrothorax and hydroperitoneum, the decrease of heart function, the results of HEstaining and VG staining all demonstrate that the model of dilatedcardiomyopathy-heart failure of rats can be made by injecting ADM into their caudalveins.2. High frequency echocardiography can exactly evaluate the heart function of themodel of heart failure caused by ADM, and the results are in high accordance with theHE staining and VG staining.3. There is a nice negative linear correlation between CVF of cardiac muscle and EF,FS.
