Objective: To isolate,purify the polysaccharides from alhagi-honey,and clarify itschemical structure.Methods:1.Water extracting-alcohol precipitating method was appliedto extract polysaccharide;The Sevag method were used to remove the Protein ofPolysaeeharide;MacroPorous adsorption resin chromatography,DEAE cellulosechromatography and Sephadex gel chromatography were applied to fractionalize andpurify.2. The homogeneity and molecular weight of every homogeneous polysaccharidewere determined by Gel filtration. The monosaccharide was identificated by Gaschromatography(GC).3. Partial hydrolysis with acid,periodate oxidation,smith degradationand NMR were used to analysis the chemical structure of polysaccharide fractions whichis high content in alhagi-honey.Results:1.Eleven Polysaeeharide fractions were got fromalhagi-honey in the first time.2. The result of Structural elucidation were as follows: themolecular weight of AP1-1was9.97×104,it was composed of arabinose,mannose,glucose,galactose,and the molar ratio was1.81∶1.10∶2.19∶4.32; the molecular weightof AP1-2was5.51×104, it was composed of arabinose,glucose,galactose,and the molarratio was1.66∶12.08∶2.92; the molecular weight of AP1-3was4.75×104,it wascomposed of rhamnose,mannose,glucose,galactose,and the molar ratio was2.11∶3.80∶4.19∶7.43ï¼›the molecular weight of AP1-4was2.75×104, it was composed ofrhamnose,arabinose,mannose,glucose,galactose,and the molar ratio was8.91∶3.29∶5.60∶0.67∶3.76;the molecular weight of AP1-5was1.85×104, it was composed ofmannose;the molecular weight of AP1-6was1.45×104,it was composed ofmannose,glucose,and the molar ratio was8.86∶7.21;the molecular weight of AP2-1was8.40×103,it was composed of rhamnose,arabinose, and the molar ratio was1.69∶17.20;themolecular weight of AP2-2was5.12×103,it was composed of rhamnose, mannose,and themolar ratio was1.43∶12.96ï¼› the molecular weight of AP2-3was4.20×103,it wascomposed of rhamnose,arabinose,mannose,glucose,galactose,and the molar ratio was10.61∶7.79∶2.54∶3.98∶1.16ï¼›the molecular weight of AP2-4was3.80×103, it wascomposed of arabinose, glucose,galactose,and the molar ratio was1.62∶9.13∶7.29ï¼›the molecular weight of AP2-5was2.83×103,it was composed of arabinose,mannose,glucose,galactose,and the molar ratio was6.83∶8.57∶10.22∶1.78.3.The main chain ofAP1-1is mainly made up ofâ†'6)-α-D-Glcp-(1â†'andâ†'2,6)α-D-Galp-(1â†', The side chain iscomposed of α-D-GalpA-(1â†',â†'5)-α-L-Araf-(1â†'and α-D-Man-(1â†'; The main chain ofAP1-3is mainly made up ofâ†'2,3)-α-L-Rhap-(1â†',â†'2,6)α-D-Manp-(1â†',â†'3)-β-D-Galp-(1â†'andâ†'3,6)α-D-Galp-(1â†', The side chain is composed ofα-D-Glcp-(1â†';The main chain of AP2-3is mainly made up ofâ†'2)-α-L-Rhap-(1â†',â†'5)-β-L-Araf-(1â†',â†'2,6)-β-D-Glcp-(1â†',â†'6)-β-D-Glcp-(1â†'andâ†'6)-β-D-Galp-(1â†',Theside chain is composed ofâ†'2)-α-L-Rhap-(1â†',â†'6)-β-D-Manp-(1â†'and β-L-Araf-(1â†'.Conclusion:1. It have been proved that the eleven polysaccharide fractions were pure andhomogeneous,so the methods we have used to isolate and purify polysaeeharides werefeasible and workable.2.Structural elucidation show that mostly of alhagi-honeypolysaccharides are complicated heteropolysaccharides with multi-branch.
