Effects Of TianXiangDan On Blood Lipid And Vascular Endothelial Growth Factor In The Arteriosclerosis In ApoE(-/-) Gene Knockout Mice

Objective:To observe the TianXiangDan on apolipoprotein E gene knockout (ApoE (-/-)) mouse in atherosclerosis (Arteriosclerosis AS) of vascular endothelial g rowth factor (vascular endothelial growth factor,VEGF)and blood lipid effects. Met hod:Sixty Eight-week old ApoE(-/-)gene knockout mice were randomly devided int o6groups:control group(fed with0.9%NACL0.2ml·d·10g-1, n=10),model group (fed with0.9%NACL0.2ml·d-1·10g-1, n=10),TianXiangDan (TXD) high dose group(fe d with TianXiangDan7.2g·kg-1·d-1n=10), TianXiangDan (TXD) medium dose group (fed with TianXiangDan3.6g·kg-1·d-1,n=10), low dose group(fed with TianXiangDan1.8g·kg-1·d-1n=10), and Atorvastatin Calcium Tablets (ATV) group (fed with ATV0.004g·kg-1·d-1n=10). The mice were sacrificed after12weeks. The lesion of athe rosclerosis was observed by microscope and pathological staining, respectively. The Plasma level of blood lipid was measured by test kit, the level of VEGF was me asured by ELISA kit. results:microscope and hematoxylinenosin staining showed t hat the area of atherosclerotic lesion was significantly less in TXD-treated groups t han in model group(P<0.05),especially TXD high dose group was the lest.The res ult of aortic arch stained showed lipid deposition was less in TXD-treated groups mice than in the model group(P<0.05), also especially TXD high dose group was the lest. Moreover, the level of VEGF was lower in TXD-treated groups mice tha n in the model group(P<0.05). also especially TXD high dose group was lowest. Conclusion:TianXiangDan could reduce the levels of serum total cholesterol (Seru m total cholesterol TC), triglyceride(Triglyceride TG) and low-density lipoprotein ch olesterol(Low density lipoprotein LDL-C) on apolipoprotein E-knockout mice. Tian XiangDan could reduce plaque of peripheral atherosclerosis in VEGF expression an d the area of AS plaque effects.
