Objective:Atherosclerosis?As?is still one of the major diseases that threaten human life and health.At present,the pathogenesis of atherosclerosis is not yet clear.In clinical studies,testosterone levels reflect androgen levels.Dihydrotestosterone?DHT?males with high dihydrotestosterone mortality rates in ischemic heart disease are reduced.Male hypotestosteroneemia is positively correlated with male atherosclerosis.Androgens mainly work through the combination of androgen receptors?AR?to form receptor ligand complexes.The role of androgens and androgen receptors on the mechanism of male atherosclerosis is not clear.In order to explore the relationship between androgens and male atherosclerosis,the research group obtained yeast myosin?Tropomyosin,TPM?3 by using yeast two-hybrid technology to screen for proteins that interact with androgen receptors in the male monocyte cDNA library,through pull down technology and western blotting technology and other in vitro protein technology to confirm the substantial interaction between tropomyosin 3 and androgen receptor.The purpose of this study was to further verify the expression of TPM3 in male mouse arterial plaques by constructing an atherosclerosis model and immunohistochemical staining.Provide theoretical basis and targets for the pathogenesis and treatment direction of male atherosclerosis.Methods:Six-week-old Apolipoprotein E gene knockout(ApoE-/-)male mice and CBA male mice were selected and sacrificed after being fed with high-fat diet for 15 weeks.Take the descending segment of arterial tissue for paraffin embedding to make wax blocks.Take the wax block of arterial tissue and observe the structure of blood vessel under HE staining microscope,whether there is plaque in the blood vessel.The wax block of arterial tissue was taken for immunohistochemical staining of tropomyosin 3 to observe the expression of tropomyosin 3 in arteries.Use Imagproplus 6.0 to analyze the picture,calculate the mean plaque thickness.To summarize the results,use Graphpad prism6 software to analyze the data and draw a statistical graph.Results:1.HE staining results show that the ApoE-/-male mouse atherosclerosis model is successfully established;2.Statistical results show that the difference between the arterial intima thickness of the ApoE-/-group and the CBA group is statistically significant?P value<0.05?;3.Immunohistochemical results showed high expression of TPM3 in the endothelium and plaques of atherosclerotic tissues.Conclusions:The high expression of TPM3 in male ApoE-/-mouse atherosclerosis tissues suggests that TPM3 is involved in the development of atherosclerosis.
