| Objective:Observe the concentration of serum E2and the expression of Bax, Bcl-2, IL-1β, MMP-13, Collagen-Ⅱ of articular cartilage,after the castration osteoarthritis rats intervened by the Guilu Erxian glue and its disassembled prescriptions at different times. Discusses the mechanism of action and the scientific connotation of the compatibility between the Monarch and his Subjects.Methods:1. Establish the castration rat model of osteoarthritis, identified by contrasting the changes of the related tissues in pathology;2. Different groups of rats are intervened by the Guilu Erxian glue and its disassembled prescriptions in2,4and8weeks;3. After the intervention of different time, take the serum and the right medial condyle femur of rats from each treatment group for the next using;4. Detect the concentration of E2, T3, T4, and Cor in serum with Radioimmunoassay;5. Detect the expression of Bax, Bcl-2, IL-1β, MMP-13, Collagen-Ⅱ in cartilage with immunohistochemistry;Results:1. The model was identified by the very significant difference between surgery and sham-operated rats in E2and glycosaminoglycan;2. The results of E2measured by RIA:2weeks:all-group were increased (P<0.05);4weeks: the full party group,Guilu group, Guilushen group, Guiluqi group were increased (P<0.05), and have no different among four groups(P>0.05);8weeks:the full party group,Guilushen group have a significantly increase(P<0.01), and Guiluqi was increased(P<0.05); GHC group have no significantly increase in each period(P>0.05);3. The results of Bax and Bcl-2expression analyzed by Immunohistochemistry:Bax:2weeks:every treating group decreased(P<0.05), Guilu group and Guilushen group were higher than the full party group and Guiluqi Group(P<0.05);4weeks:every treating group decreased(P<0.05); the four-group of TCM had no difference(P>0.05);8weeks:every treating group decreased (P<0.05). Guilu group and Guilushen group were higher than the full party group and Guiluqi Group(P<0.05); Bcl-2:every treating group increased(P<0.05) in three periods, and Guilu group, Guilushen group were lower than the full party group and Guiluqi Group(P<0.05) in three periods;4. The results of IL-1βand MMP-13expression analyzed by Immunohistochemistry:IL-1β: every treating group decreased (P<0.05) in three periods, Guilu group and Guiluqi group were higher than the full party group and Guilushen Group(P<0.05) in three periods; in8weeks the full party group and Guilushen group had no difference with GHC group; MMP-13:2weeks: every treating group decreased(P<0.05) except Guilu group;4and8weeks, every treating group decreased(P<0.05), Guilu group and Guiluqi group were higher than the full party group and Guilushen Group(P<0.05);5. The results of Collagen-II expression analyzed by Immunohistochemistry:2weeks, every treating group increased (P<0.05) and have no difference(P>0.05);4and8weeks, every treating group increased (P<0.05), the full party group was higher than others(P<0.05), Guilu group was lower than others(P<0.05);Conclusions:1. Guilu Erxian Glue and its disassembled prescriptions can improve the estrogen of rat model,Guilushen played the first important role;2. Guilu Erxian Glue and its disassembled prescriptions have no effect on Cor, T3, T4;3. Guilu Erxian Glue and its disassembled prescriptions can inhibit the apoptosis of chondrocyte, and Guiluqi played the first important role;4. Guilu Erxian Glue and its disassembled prescriptions can inhibit the local inflammation of the model, and Guilushen played the first important role;5. Guilu Erxian Glue and its disassembled prescriptions can increase the content of Collagen-Ⅱ in cartilage matrix of the model, and the full party group can do better;6. Guilushen group can improve E2better;Guiluqi group can inhibit the apoptosis of chondrocyte better; The full party group can increase the content of Collagen-Ⅱ in cartilage matrix of the model better. These may be one of the mechanisms of the compatibility. |
