Effect Of Guilu E Rxian Glue And Its Separate On Bone Resorption Of Culturing Osteoclasts

Objective:Observe how the Guilu Erxian Glue affect the bone resorption of resorption of culturing osteoclasts.In accordance with the principle of primary and secondary, divided into three groups of GUILU,GUILUSHEN,GUILUQI,by observing them how to effect the TRAP,CTsK,CTR. Compare them how to affect the bone resorption of culturing osteoclasts, compare the Guilu Erxian Glue and its Separate. And explore the significance of the Guilu Erxian Glue primary and secondary collocation.Methods:1.To prepare containing serum of Guilu Erxian Glue and its Separate.2.To establish a vitro culture system of osteoclasts.3.Prepare the medicated serum of GUILUERXIAN Glue and its Separate.4.To observe the expression of gene TRAP,CTsK and CTR in the culturing osteoclasts through Guilu Erxian Glue and its Separate containing serum’s intervention by RTF-QPCR.Results:The containing serum of Guilu Erxian Glue and its Separate can reduce the proliferation of TRAP and CTsK,promote the proliferation of CTR.The gene of TRAP intervention by RTF-PCR showed showed that:the group of Guilu Erxian Glue have the best effect, others also have significant difference;The efficacy of Guilu Erxian Glue group is stronger than the GUILU group and GUILUSHEN group. From RT-PCR result,we can find each group containing serum,in a certain extent, can reduce the expression of TRAP.The efficacy of Each group containing serum of inhibition of CTsK expression and inhibition as follows:Guilu Erxian Glue>GUILUSHEN>GUILUQI. PCR conclusion that each group containing serum can be a certain degree of promote the proliferation of CTR, the best effect comes from the Guilu Erxian Glue group others have certain significant difference,the results as follows:Guilu Erxian Glue>GUILUSHEN>GUILUQI.Conclusion:Guilu Erxian Glue and its Separate can reduce the proliferation of TRAP,CTsK and promote the proliferation of CTR of culturting osteoclasts.Generally, in reducing the expression of TRAP of culturting osteoclasts, Guilu Erxian Glue is the best,it also shows that the function of Renshen and Gouqi are quite familiar. In reducing the expression of CTsK of culturting osteoclasts, Guilu Erxian Glue is the best,it also shows that the function of Renshen and Gouqi are quite familiar.in promoting the expression of CTR of culturting osteoclasts, Guilu Erxian Glue is the best,it also shows that the function of Renshen and Gouqi are quite familiar. GuiBan formula and LuJiao formula educed the leading fuction in the whole formula,they are the major drugs in the whole formula.
