| The prescription of "BiTongtie" comes from a famous doctor. According to the basic pathogenesis of qi stagnation and blood stasis, the doctor developed a related BiTongtie after accumlating clinical practice for a long time. BiTongtie is a compond TCM of external application which has efficacy of activiating blood, dissolving stasis and analgesia. The characeristics of BiTongtie is definite curative effect and convenient operation, which is used for tereating soft tissue injury.Objective:To study on pharmacological action of BiTongtie which is used for acute soft tissue injury with the model for animal experiment, it aimed to explore and further clarify its mechanism of action, and look forward to provide scientific basis for clinical reasonable using of medicine.Methods:This subject mainly observed the pharmacological action of BiTongtie treating acute soft tissue injury by four aspects as following:(1) To observe the rat and rabbit model of acute soft tissue injury by heavy hit for several times on the same place. The effects of bitongtie were observed on injury signs index, tissue histopathological, injured focal tissue PGE2and IL-1βon the first day, the fourth day and the seventh day.(2) Promote blood circulation to remove blood stasis experiment, according to the experiments of the clotting time of mice, carrageenan induced mouse tail thrombosis and the microcirculation of normal mice and stasis type mice to observe the promotion of blood circulation of the drug.(3)Anti inflammatory experiments, according to the experiments of xylene-induced ear edema in mice, carrageenan induced rat paw swelling, acetic acid-induced abdominal capillary permeability and cotton pellet granuloma to observe the anti-inflammatory effect of drug.(4)Analgesic experiments,according to the the experiments of acetic acid writhing test, the hot plate test and formaldehyde induced mouse pain to observe the analgesic effect of drug.Results:(1) In rat model of acute soft tissue injury, compared with the model group and vehicle group, the middle dose and high dose of bitongtie can improve the injury signs index (P<0.05, P<0.01), tissue histopathological on the fourth day, the seven day after making model; at the same time the different dose of bitongtiecan reduce the content of PGE2and IL-1β (P<0.05, P<0.01). In rabbit model of acute soft tissue injury, compared with the model group and vehicle group, the middle dose and high dose of bitongtie can improve the injury signs index, tissue histopathological on the fourth day, the seven day after making model; at the same time the different dose of bitongtiecan reduce the content of PGE2and IL-1β (P<0.05).(2) Promote blood circulation to remove blood stasis experiment. Compared with the model group and vehicle group, BiTongtie has no effect on the clotting time of mice, however, can reduce the carrageenan induced thrombosis length and each group can notably increase the earvenule, arteriole diameter, the blood flow speed of normal mouse and the mouse ear microcirculatory disturbance induced by adrenaline(P<0.05. P<0.01).(3) Anti inflammatory experiments, the high dose of BiTongtie can obviously reduce the xylene-induced ear edema in mice (P<0.01). the middle and high dose of BiTongtie can inhibit the rat paw swelling (P<0.05), decrease abdominal capillary permeability (P<0.05), reduce the weight of granuloma (P<0.05).(4) Analgesic experiments.the high dose of BiTongtie can prolong the pain latency and reduce the writhing times, it can also decrease the second phase (10min-25min) pain score (P <0.05), however, the drug has no effects on hot plate test.Conclusion:BiTongtie has efficacy of activiating blood, dissolving stasis and analgesia, which can improve the swelling, blood stasis, analgesic of soft tissue injury. The durg can decrease the content of PGE2and IL-1β, improve the tissue histopathological. As a result, BiTongtie has the clear effect on the soft tissue injury. |
