Objective:To detect the expression and correlation of Wnt5a, β-catenin and c-myc mRNA and the expression Wnt5a, P-catenin and c-myc protein were also examined in human breast carcinoma and breast fibroadenoma tissue.Approach the relationship between expression of wnt5a, P-catenin and c-myc protein and clinic pathological factors and discuss the correlation with the expression of wnt5a, P-catenin and c-myc in order to explore their mechanism of molecularbiology for the occurrence and development of breast carcinoma. Methods:Wnt5a, P-catenin and c-myc mRNA and protein were detected by RT-PCR and SP irnmunohistochemical method in24breast carcinoma tissues and24normal breast tissues and20breast fibroadenoma tissues. The results was treated by matehed-pairs T test, chi-square test, Fisher ecact test and spearman correlation analysis.Results:①The expression of Wnt5a、β-catenin、c-myc gene were detected in breast carcinoma tissues and breast fibroadenoma tissues respectively;Wnt5a mRNA relative content in breast carcinoma tissues and breast fibroadenoma tissues was0.5183±0.21235,0.3355±0.15035respectively; P-catenin mRNA relative content in breast carcinoma tissues and breast fibroadenoma tissues were0.5467±0.12324,0.3230±0.06317 respectively; c-myc mRNA relative content in breast carcinoma tissues and breast fibroadenoma tissues were0.4449±0.16586,0.3115±0.09385respectively. The exprssions of Wnt5a、β-catenin、c-myc gene among breast fibroadenoma tissue and breast carcinoma tissue showed significant difference (P<0.05)②The positive expression rate of Wnt5a protein in breast carcinoma tissues and normal breast tissues and breast fibroadenoma tissues was83.3%(20/24cases),33.3%(8/24cases),50.0%(10/20cases) respectively. The positive expression rate of Wnt5a protein in breast carcinoma tissues was sharply higher than normal breast tissues and breast fibroadenoma tissues, the positive expression rate of Wnt5a protein among breast fibroadenoma tissues,normal breast tissues and breast carcinoma tissues showed significant difference (P<0.05);③The expression of Wnt5a、β-catenin、c-myc protein in breast carcinoma tissues was not associated with any significant relation to patient age、tumor size、lymphnode metastasis、histological grade and clinical stage(P>0.05).④Wnt5a protein did not show correlation with P-catenin protein and c-myc protein(P>0.05).The expression of Wnt5a, β-catenin and c-myc in the breast carcinoma tissues didn’t show any significant relation to the presence or absence of ER, PR, P53, CerbB-2, VEGF,and ki-67(P>0.05). Conclusions:The Wnt5a, β-catenin andc-myc gene may have been actived in period of breast carcinoma, Wnt5a may become a molecular marker and may cancer gene stimulator function in breast carcinoma.
