Objective:To investigate the expression of Wnt5 a and β-catenin in human Endometrial adenocarcinoma(EAC) and to analyze their relationship with the biological characteristics of malignant, clinical stage, pathological grade, and invasion and metastasis of EAC and clinical significance.Methods:Fifty-six cases tissues of EAC were collected from the First Affiliated Hospital of Henan University of Science and Technology from 2007 to 2009. The Wnt5 a protein and β-catenin membrane protein in normal endometrium(13 cases), endometrial atypical hyperplasia group(18 cases), endometrial adenocarcinoma group(56 cases) was detected by immunohistochemical SP technique. According to the clinical stage FIGO2009 method: there were phase I ~ II 38 cases, 18 cases III ~ IV; 18 cases of lymph node metastases. All specimens were identified by histopathologic, and the patients didn’t perform radiotherapy and chemotherapy before surgery. All specimens were fixed in 10% formalin solution, embedded in paraffin. The expression of Wnt5 a and β-catenin in 56 cases of EAC, 13 normal proliferative phase endometrial tissue,18 typical endometrial tissues, were detected by Immunohistochemical SP method,and we analyed their relationship with the malignant biological characteristics of EAC and their correlation. SPSS16.0 software was used for data analysis, χ2 test was used to compare the expression rate between the two sample, whereas the correlation between the two variables was determined using the Spearman rank correlation analysis,and Kaplan-Meier Survival Analysis,P<0.05 was indicated statistical significance.Results:The positive expression of Wnt5 a protein located in cytoplasm, as tan particles, the expression of Wnt5 a in 56 intrauterine membrane adenocarcinoma examples was significantly lower than endometrial atypical hyperplasia and normal breeding period endometrial tissue, there were significant difference between three groups(P<0.05), there was differences between the expression of Wnt5 a in endometrial adenocarcinoma and the pathological grading(P<0.05), however, there was no difference between clinical stages and lymph node metastasis(P>0.05). The positive expression of β-catenin/cytoplasm located in cell membrane as tan particles, The expression of β-catenin in 56 intrauterine membrane adenocarcinoma examples was significantly higher than in endometrial atypical hyperplasia and normal endometrium tissue of expression, and there were statistically significant difference(P <0.05), There was differences betweeen the expression of β-catenin in endometrial adenocarcinoma and the pathological grading(P<0.05), however, there was no difference between the clinical stages and lymph node metastasis(P>0.05). Expression of both types of has nothing to do with patient’s age, there was no statistically significant difference(P>0.05). In endometrial adenocarcinoma tissues, it showed that the expression of Wnt5 a and β-catenin was significantly negative correlation using the Spearman rank correlation analysis(rs=-0.638, P<0.05). Kaplan and Meier survival analysis showed that Wnt5 a express positive cases overall survival rate was higher than the negative case, difference was statistically significant(P=0.024), β-catenin expression positive cases overall survival rate is lower than the negative case, difference was statistically significant( P=0.044).Conclusion:1. Positive expression of Wnt5 a in endometrial adenocarcinoma group is lower than in the normal period of endometrial proliferation and atypical hyperplasia of endometrium. Positive expression of β-catenin expression in endometrial adenocarcinoma group is higher than the normal period of endometrial proliferation and atypical hyperplasia endometrium group.2. Expression of Wnt5 a and β-catenin in endometrial adenocarcinoma was associated with a pathologic stage, and there was nothing to do with the age, clinical stage and lymph node metastasis. the results indicated that Wnt5 a and β-catenin may play an important role in development and incidence of endometrial cancer.3. Wnt5 a and β-catenin mutual antagonism, jointly participate in the occurrence of endometrial cancer development, joint detection of two factor may have on the recurrence and prognosis of endometrial adenocarcinoma detection feature which provides the basis for judgment.
