Objective:Polyphenolic active ingredients were extracted from banana peel,which can effect on the hyperlipidemic and atherosclerotic rats, to explore its hypolipidemic and antioxidative mechanism. And after the rats were sacrificed,their aorta was dissected to made pathologic slices, to further explore the hypolipidemic and anti-atherogenic mechanism of banana peel polyphenols.Methods:1.Microwave-assisted extraction of banana peel polyphenols, freeze-dried purified, spare2.Sixty healthy SPF grade male SD rats were randomly divided into six groups,each contained10rats:normal control group (NC), model control group (MC), positive control group (PC, simvastatin group),the low dose group of banana peel polyphenols (PI), the dose group of banana peel polyphenols(P2), the high dose group of banana peel polyphenols(P3), after the adaptability of feeding for a week.To made high fat diet model and be the experimental intervention with a banana peel polyphenols, the weekly said a body weight of rats.After feeding12w, fasting12h,collect the orbital venous blood serum for the tests of TG, TC, HDL-C and LDL-C,and the activities of SOD, MDA,T-AOC.3.At the end of the experiment, rats were killed by Cervical dislocation and immediately stripped the aorta fixed with10%formalin, paraffin embedded, sectioned, and stained by HE and MMP-9immunohistochemical staining to observe the organizationchanges of the aorta under the light microscopeResults:1.Weight changes of rats in each group before and after the intervention:when the experimental went forl2w, compared with NC group, the consumption of high fat diet body weight of rats were significantly lower (P <0.05), and compared with the MC group, the PC P2and P3groups’weight gained obviously(P<0.05)2.Lipid changes:when the experimental went forl2w, compared with NC group, TG,TC, LDL-C value of MC group rats was significantly increased (P <0.05), HDL-C value was significantly lower (P<0.05). Compared with the MC group, the banana peel polyphenol intervention can significantly reduce TG, TC and LDL-C values (P<0.05) and increase in HDL-C values (P<0.05).Vasoactive substances changes:when the experimental went for12w,compared with NC group, the serum SOD, T-AOC of MC group was significantly lower and MDA significantly higher (P<0.05). Compared with the MC group,after the banana peel polyphenol intervention the activities of SOD, T-AOC was significantly increased, MDA decreased significantly (P<0.05)3.HE staining and MMP-9immunohistochemical staining:Compared with NC group, MC group rats had vascular intimal thickening, lipid deposition and foam cell formation, and liver steatosis; MMP-9was expression in AS plaques. Compared with the MC group, Banana peel polyphenol intervention groups were mild lesions, MMP-9expression, but the positive rate was not high. Conclusions:1. Banana peel of polyphenols were taken by hyperlipidemic rats with lipid-lowering and antioxidant effects2. Banana peel polyphenols could reduce damage to the arteries through the lipid-lowering antioxidant mechanism, thus delaying the development of the AS3. Propylthiouracil in the high fat diet could lead to low thyroid function in rats,which is conducive to the formation of hyperlipidemia and could cause the rat growth retardation, low birth weight in the experimental conditions.
