Objective:To observe the effect of pomegranate peel polyphenols ointment on HIF-1?/IL-17 signaling pathway in acne-like skin lesion model of rats,and to explore the anti-inflammatory effect and mechanism of pomegranate peel polyphenols on acne.Methods:36 healthy without specific pathogen(SPF)level in rats according to random number table,on average,divided into blank group,model group,low pomegranate peel polyphenols in high concentration group,positive control group,except the blank group,the rest of the group in the rat ear side ear catheter opening evenly 100%oleic acid,0.5 ml/second,1 time/day,and 21 days with oleic acid began in the same part rats auricle subcutaneous injection for three days the acne propionic acid bacillus liquid 50 mu,1 1 time/day,rats auricle acne model was constructed,after the success of the macroscopic observation suggests building external use drugs,The pomegranate rind polyphenol group was given 0.5g pomegranate rind polyphenol ointment of different concentrations,the positive group was given 0.5g clindamycin hydrochloride gel,and the blank group and model group were given the same amount of distilled water,twice a day,for 2 consecutive weeks.24h after the last administration,tissues from the auricle model of rats were taken and the changes of skin lesions in each group were observed and recorded.The pathological changes of skin in each group were observed by HE staining.IL-17 level in serum of rats was detected by ELISA method,and the expression levels of mTOR,HIF-1? and ROR?t in local tissues were detected by immunohistochemistry method.Results:1.The naked eye observation showed that compared with the model group,the skin lesion surface of the auricle model of rats in the pomegranate peel polyphenol group and the positive control group was smoother,the epidermal keratosis was significantly improved,and no papular-like cyst or dandruff were observed.2.HE staining showed that,compared with model group,epidermal keratosis was improved in pomegranate peel polyphenols medium and high concentration groups,and the infiltration of inflammatory cells in epidermis and dermis was reduced;In the positive control group,there was no obvious enlargement of hair follicle and funnel,the epidermis was significantly improved,the thickness of spinous layer and granular layer was reduced,and the infiltration of inflammatory cells in epidermis and dermis was significantly reduced.3.ELISA test showed that compared with blank group,the levels of IL-17 inflammatory cytokines in model group,pomegranate peel polyphenol(low,medium and high)concentration groups and positive control group were significantly increased,with statistical significance(P<0.05).Compared with model group,IL-17 level in pomegranate peel polyphenols(low,medium and high)concentration groups and positive control group was significantly decreased,with statistical significance(P<0.05).Compared with the low concentration pomegranate peel polyphenol group,the IL-17 content of high concentration pomegranate peel polyphenol group was significantly decreased,and the difference was statistically significant(P<0.05).4.Immunohis to chemical assay showed that compared with blank group,HIF-1? decreased significantly in pomegranate peel polyphenols low and medium concentration groups and positive control group,with statistical significance(P<0.05).Compared with model group,HIF-1? decreased significantly in pomegranate peel polyphenol low,medium and high group and positive control group,and the difference was statistically significant(P<0.05).Compared with model group,ROR?t in pomegranate peel polyphenols low and high concentration groups was significantly decreased,with statistical significance(P<0.05).There was no significant difference in mTOR expression among all groups(P>0.05).Conclusion:1.Pomegranate peel polyphenol cream can significantly inhibit inflammation and treat acne.2.Pomegranate peel polyphenol cream can inhibit the expression of HIF-1?/IL-17 signaling pathway,inhibit the inflammatory response,and then play a role in the treatment of acne.
