Objective:To investigate the effect of gastrodia ELATA (Gastrodia ELATA B1, GE) and gastrodine (its main ingredient) on the repair of peripheral nerve regeneration, to lay an experimental foundation in expanding the range of indications and administration of gastrodia ELATA Blume, to provide new choice of clinical drugs on nerve repair and regeneration, and to further clarify the effect and mechanism of rhizoma gastrodiae on peripheral nerve function.Methods:Animal experiment:40rabbits were randomly separated into four groups, the blank control group (group A), Gastrodin injection group (group B), Local muscle injection group (group C) and Gastrodia elata powder filling stomach group (group D). These rabbits were feed in different cages by specialists. To each individual, the left thigh sciatic nerve was cut off and then sewed up to make the model of sciatic nerve totally injury while contralateral was used as control. The rabbits in groups B and C were daily vein-giving medicine or local muscle-giving medicine, In group D, gastrodia was firstly milled, and then refined by100mesh, lastly mixed into paste by water and transferred into stomach. The survival rates in each group were observed during the feeding process, at t=4,8and12weeks the nerve conduction velocity was measured by nerve electrophysiology while the specimens were observed by appearance, light and electron microscop. At the same time, the target muscle (triceps muscle) was assayed for wet weight. Take the target muscle (triceps surae) specimens, detection of wet weight. Because all groups showed a normal distribution, the intergroup differences were assessed using parametric statistical methods, a one-way paired independent sample of q-tests.Results:The results are analyzed as follows.①nerve conduction velocity:the nerve conduction velocity at t=4weeks, muscle group, the intravenous group or gavage group is faster than the blank group, but there is no significant difference (P>0.05) at t=8weeks, the MNCV muscle group, the intravenous group or gavage group was better than the blank group, the difference was significant (P<0.01, P<0.01, P<0.01). Gavage group is better than the intravenous group or muscle groups with significant difference (P <0.05, P<0.05) while there is no significant difference between muscle group and intravenous group (P>0.05) and there is no significant difference between muscle group and intravenous group (P>0.05); at t=12weeks, the MNCV muscle group, the intravenous group or gavage group was better than the blank group after12weeks with significant difference (P<0.01, P<0.01, P<0.01). The conduction velocity in gavage group is better than the intravenous group or muscle group with no significant difference (P>0.05, P>0.05) while there is no significant difference between muscle group and intravenous group (P>0.05).②The recovery rate of wet weight of muscles:at t=8weeks, the recovery rate of muscle group, the intravenous group or gavage group was better than the blank group with significant or very significant difference (P<0.01, P<0.01, P<0.01), and there is no significant difference between muscle group and intravenous group (P>0.05), between muscle groups and gavage group (P>0.05), or between the intravenous group and oral group (P>0.05); So is it at t=12weeks.③Immunohistochemistry:CD34staining is used to observe the number of microvessels. Att=8weeks, MVD counting muscle group, the intravenous group and the gavage group are better than the blank group with significant difference (P<0.05, P<0.05, P<0.05). There is no significant difference between muscle group and intravenous group (P>0.05), between muscle group and gavage group (P>0.05), or between intravenous group and gavage group (P>0.05); So is it at t=12weeks.④The detection of nerve’s ultrastructure:at post-surgery12weeks, nerve’s ultrastructure in the anastomotic distal of each group was observed, The electron microscopy showed that gavage group, the intravenous group and muscle group had more the regenerated nerve fibers and the nerve fibers were relatively thick, the myelin sheaths were nearly normal with thick, compact and distinct lamellar; Blank group had less regenerated nerve with irregular shape, obvious deformation, thinner myelin sheaths, partially separated lamellar, less maturity and scattered regenerative unmyelinated fibers.Conclusions:The results of experimental observations and data analysis showed that:①Gastrodia played a somewhat active role in the repair and regeneration of peripheral nerve.②The gastrodin, the main extracts from gastrodia, played an active role in the repair and regeneration of peripheral nerve.③The Gastrodia powder and gastrodin have no significant difference in promoting the growth of the peripheral nerve. Therefore, a certain amount of Gastrodia or gastrodin is able to promote the regeneration and repair of the peripheral nerve to some degree.
