Objective:During the the process from onset to progression of Carotid atherosclerosis, there area variety of immune inflammatory cells involved in the mechanism. And because of theimmune inflammation of lesions area, the cerebrovascular disease and vascular events willcontinue to happen. Dendritic cells play an important role in the immune mediatedinflammation response, In this study, there are different study groups according to thecarotid atherosclerosis plaque nature and intima-media thickness of the object, and withflow cytometric detection of peripheral blood dendritic cells molecular marker CD11c andCD86. Then, these data will be relevant comparison with the lipids, uric acid andC-reactive protein and other clinical data. And then to explore the role and mechanismof the occurrence and the pathogenesis of DC and other immune inflammatory cells inthe carotid artery atherosclerosis.Methods:The experiment selected a total of108cases as the object of the patients with carotidatherosclerosis and the healthy people from December2010to December2011in TaishanMedical University affiliated hospital.Carotid atherosclerosis patients with cerebrovascularsymptoms and signs are78cases. In the same period,30individuals who is healthy as acontrol group. All objects by color Doppler ultrasound detection of carotid arteryatherosclerosis and check the brain MRI, blood lipids, blood glucose, blood pressure,C-reactive protein, uric acid, and record the clinical data of all selected objects. Thediagnosis of our patient group at the same time meet the diagnostic criteria of the varioustypes of cerebrovascular disease. We have a person responsible for the detection of carotidatherosclerotic plaque in echo properties and intima-media thickness, we used flowcytometry analysis of peripheral blood mononuclear cells directly labeled mouse anti-human monoclonal antibody staining of CD11c~+and CD86~+cells, and then comparethe CD11c~+CD86~+double positive cells accounted for the proportion of peripheral blood mononuclear cells. Through this study to investigate the inflammatory immune functionand mechanism of dendritic cells in the pathogenesis of carotid atherosclerosis. Use SPSS16.0statistical software to analysis the data.Results:1. Comparison of the proportion of CD11c~+CD86~+double positive cells withdifferent plaques groups: the proportion of CD11c~+CD86~+double positive cells wereincreased in low echo group, mixed echo group and strong echo group compared withcontrols, difference is statistically significant (P<0.01). the proportion of CD11c~+CD86~+double positive cells in the unstable plaque group were increased compared with in thestable plaque group, the difference was statistically significant (P<0.01). the proportion ofCD11c~+CD86~+double positive cells in low echo group were increased compared with inmixed echo group, the difference was statistically significant (P<0.05).2. Comparison of the proportion of CD11c~+CD86~+double positive cells withdifferent intima-media: the proportion of CD11c~+CD86~+double positive cells in group of0.9≦IMT﹤1.0, group of1.0≦IMT﹤1.1and group of1.1≦IMT﹤1.2were increasedcompared with controls, difference is statistically significant (P﹤0.01). the proportion ofCD11c~+CD86~+double positive cells in group of1.1≦IMT﹤1.2and group of1.0≦IMT﹤1.1was increased when compared with group of0.9≦IMT﹤1.0, difference isstatistically significant (P﹤0.01). There was no significant difference between group of1.0≦IMT﹤1.1and group of1.1≦IMT﹤1.2(P﹥0.05).3. The proportion of CD11c~+CD86~+double positive cells and the levels of totalcholesterol are positively correlated (P=0.002,r=0.295). The proportion of CD11c~+CD86~+double positive cells and the levels of low-density lipoprotein are positivelycorrelated (P=0.001, r=0.318). The proportion of CD11c~+CD86~+double positive cellsand the levels of high-density lipoprotein are negatively correlated (P=0.018, r=﹣0.228).The proportion of CD11c~+CD86~+double positive cells and the levels of Systolic bloodpressure are positively correlated (P=0.031, r=0.248).4. The proportion of CD11c~+CD86~+double positive cells and the levels of blooduric acid are positively correlated (P=0.001, r=0.304).5. The proportion of CD11c~+CD86~+double positive cells and the levels of C-reactive protein are positively correlated (P=0.042, r=0.315).Conclusion:1.Carotid atherosclerosis in the occurrence and stability changes when the molecules of the dendritic cell surface marker CD11c and CD86appeared to change, which play arole in the start of immune inflammation and the pathogenic process.2. The CD11c+CD86+double positive cells are related to the body’s lipids(CHOL, LDL and HDL), systolic blood pressure, uric acid, C-reactive protein.These riskfactors.may be involved in dendritic cell maturation and carotid atherosclerosis pathogenicprocess.
