Phyllanthus emblica L. is a member of Euphorbiaceae family. There are nearly16ethnics such as Han, Tibetan, Dai using this kind of medicine which is embodied in hinese pharmacopoeia. It’s used in the treatment of blood disease, the Tripa disease and Bacon disease as a Tibetan medicine named "Jerura"."Triphala" is a compatibility of fructus phyllanthi, fructus chebulae and fructus terminaliae billericae. In this paper, we studied on HPLC-FPS, determination method and the anti-oxidant activity in vitro of polysaccharide with high-performance liquid chromatography, UV-visible spectrophotometry.Objective:1. Establish the HPLC-FPS and develop an RP-HPLC method for the determination of gallic acid,corilagin and ellagic acid in Phyllanthus emblica.2. Study on the antioxidant activity in vitro and develop an method for the determination of polysaccharide in Phyllanthus emblica.Methods:1. HPLC analysis was performed on a Ultimate LP RP-C18(4.6mm X250mm,5μm, Welch) column eluted with acetonitrile(A) and0.1%Phosphoric acid solution(B) in gradient elution. The flow rate was0.8mL·min-1. The column temperature was20℃and the detection wavelength was220nm.2. Reserch the anti-oxidative effect of polysaccharide with DPPH·test.3. Determinaton of the content of polysaccharide by phenol-sulfuric acid method and UV-vis spectrophotometric.Results:1. The gallic acid content of each batch between1.15%-1.78%, the corilagin content of each batch between0.43%-0.78%, the ellagic acid content of each batch between0.64%-1.14%, the polysaccharide content of each batch between14.22%-20.79%.2. There are14common peaks in total, calculating similarity degree by vector included-angle cosine method, and using correlation coefficient method to confirm result, both above0.90.3. The half maximal inhibitory concentration of polysaccride for DPPH·was0.211mg·mL-1.Conclusion:1. The HPLC method and UV-vis spectrophotometry method are simple, accurate,repeatable and stable, which can be used for quality control of Phyllanthus emblica.2. Phyllanthus emblica have certain anti-oxidant activity, the scaventing capacity for DPPH·increases with the concentration, and its action is weaker than vitamin C and galic acid.
