Syringa Reticulata belongs to Oleaceae family and Syringa Linn genus.The whole plant have an good effect on remedying. The research mainly on chemical constituents, antioxidant and antimicrobial activities of Syringa Reticulata, and optimize the extraction process for active compounds in it.The research provides a theoretical basis for the better development and utilization of Syringa Reticulata resources.Review the literatures of chemical constituents and pharmacological actions of plants belongs to Syringa Linn at the beginning of the study to research Syringa Reticulata deeper.The chemical constituents research is systematically. Syringa Reticulata were ultrasonic extracted with the10%of the ethanol. Concentrate the extract and pass it through D101microporous resin. Remove the pigment and impurities with water and obtained the10%,30%,50%ethanol parts.Concentrate every part respectively.7compounds were isolated from the10%and30%ethanol parts by normal and inversed phase chromatography.4compounds were identified as D-mannitol, syringin,(8E)-ligstroside and oleuropein by physical and chemical properties, spectroscopic methods(MS,1H-NMR,13C-NMR) and literatures.Analyze the essential oil of Syringa Reticulata Bark by GC-MS.46peaks were isolated by capillary column, of which28compounds were identified. Analyze the relative content of each constituent, and the result suggests that9,12-Octadecadienoic acid (Z,Z)(28.52%), palmitic acid (27.46%),9,17-Octadecadienal(Z)(14.16%), Tricosane (2.44%) have the higher content.As the result show us, Syringa Reticulata Bark essential oil is rich in fatty acids, alkanes, alcohols and ketones.Compare the syringin content of Syringa Reticulata Bark in different collecting time by HPLC. The syringin content is highest in April16th (1.772%) and decline in May and June,and it down to the lowest in May21st(0.130%), but raise again in September. The results suggest that the best collecting time of Syringa Reticulata Bark is mid-April and late September.Optimize the syringin extraction process in Syringa Reticulata Bark according to central composite design and response surface methodology. The main influential factors of extraction efficiency are reflux time, ethanol concentration and solvent ratio. The conditions of the extraction were evaluated by the total content of syringin. Multiple linear regression and binomial fitting models were performed with the main influential factors. Indicate the optimum extracting technology of syringin are46%ethanol,20fold solvent and ultrasonic extraction80minutes. This method is more convenient and with an higher precision.Determine the antioxidant activity of the extract in vitro by three methods including DPPH assays, ABTS assays and reducing power. The result suggests that the total extract, the10%part and the30%part have the more obvious effect than others. The30%part works best, and it can scavenge DPPH and ABTS radicals as efficient as Vc when the concentration reach0.150mg/mL and0.620mg/mL respectively.The antimicrobial activities of Syringa Reticulata Bark extract and essential oil were analyzed by the filter papers and show vary results of against5kinds of bacteria. The extract have the better effect to Staphylococcus aureus and Pasteurella, and the effects are stable under heating, pressure, ultraviolet radiation and acidic condition. The essential oil against Pasteurella, Escherichia coli, Salmonella enterica better.Innovative features1.Analyze the essential oil of Syringa Reticulata Bark for the first time. The result suggests that Syringa Reticulata Bark essential oil is rich in fatty acids, alkanes, alcohols and ketones.2.Optimize the syringin extraction process in Syringa Reticulata Bark based upon chemical constituents research,and provide a theoretical basis for the industrial production in the future.3. The bark of Syringa total extract and different concentrations of macroporous resin alcohol site in vitro antioxidant activity were studied and screening.The research can provide the theoretical basis to natural antioxidant.4.The antimicrobial activities of Syringa Reticulata Bark extract and essential oil were analyzed by the filter papers and show vary results of against5kinds of bacteria(Staphylococcus aureus, Pasteurella, Escherichia coli, Salmonella enterica, Streptococcus).The research can provide the theoretical basis to natural antimicrobials.
