ObjectiveApplication MSCT Perfusion technology to explore the relation betweenthe rabbit VX2planting tumor emboli portal vein infusion parameters and theexpression of the VEGF,in order to lay the foundation of portal venous tumoremboli further experimental study.MethodsSelected13healthy Japanese white rabbits, taked1only for the passageof tumor inoculation,another12as experimental rabbits; Thawed VX2tumorlines Preparation of tumor cell suspension and injected in the medial area ofthe muscles of the hind shares of Japanese white rabbits, taked tumor from theinjection site after two weeks, the formation of solid tumors were removed bysterile surgical, cuted the strong growth of the tumor edge fish tissue, cutedinto thin strips structure and prepared for cultivation of tumor cells spare,randomly selected from six rabbits which were anesthesiaed, epilationed,disinfectioned, after surgical laparotomy, Punctured the superior mesentericvein tumor strain Organization and injected into the portal vein as rabbit portalvein planted VX2tumor thrombus in animal model; the remaining six rabbitsafter caesarean section and punctured mesenteric vein then Saline injectedinto the portal vein as the control group.Observed portal vein tumor thrombus growth by16-slice spiral CT enhanced scan after2weeks, Determined portal vein tumor thrombusunderwent CT perfusion, Scanned data through the LAN to ADW4.2workstation, used GE’s Perfusion3body tumor perfusion softwaredeconvolution model for data processing, Measured portal vein tumorthrombus blood flow(HBF)、mean transit time(MTT) and the change ofcapillary permeability surface. Executioned the rabbits after scanning andRespectively removed the transplanted VX2tumor thrombus of the portal veinunder sterile surgical conditions. Under the removal of the portal veintransplant VX2tumor thrombus organization did immunohistochemicaldetection of VEGF expression.Respectively compared the average of tumor block,non-tumor livertissue, normal liver perfusion parameters(HBF、 HBV、 MTT、 PS), All datawith the statistical package were trated by SPSS17.0, Measurement datawere expressed as mean±standard deviation, reference range applied95%confidence interval, used independent t-test between the two groups, therewere significant differences with P<0.05portal vein tumor thrombus CTperfusion values and expression of VEGF in the amount of correlation analysisapplied Spearman rank correlation analysis, there were significant differenceswith P<0.05.Results1MSCT liver perfusion HBF parameters, HBV and PS of the non-tumoremboli area from the experimental group compared to normal control group,the difference was not statistically significant (P>0.05).2The experimental group with portal vein tumor thrombus compared to non-tumor emboli area and normal control group respectively, rabbit VX2tumoremboli edge MSCT perfusion HBF parameters, HBV and PS heighten, thedifference was statistically significant (P<0.05).3The experimental group with portal vein tumor thrombus compared to non- tumor emboli area and normal control group respectively, rabbit VX2tumoremboli fringe area mean MTT was reduced, the difference was statisticallysignificant (P<0.05).4MSCT perfusion HBF, HBV and PS parameters mean of Rabbit VX2tumoremboli and the expression of rabbit VX2tumor emboli had positive relation(P<0.05),but MTT and the expression of VEGF had negative relation(P<0.05).ConclusionsMSCT perfusion parameters of rabbit portal vein VX2planting tumoremboli and the expression of VEGF had relation, which could evaluate theangiogenesis of portal vein VX2planting tumor emboli.
