ObjectivesTo investigate the polymorphism of15STR (short tandem repeat)loci(D8S1179, D21S11, D7S820, CSF1P0, D3S1358, D5S818, D13S317,D16S539, D2S1338, D19S433, vWA, D12S391, D18S51, Amelogenin,D6S1043, FGA) in Mongolian Ethnic Group of Snow Mountain. So as to revealthe genetic relationship among Mongolian Ethnic Group in Snow Mountain andother29domestic ethnic groups. The study is an evidence of this nationalityorigin in genetics.MethodsSpecimens of blood cakes form198unrelated Mongolian individuals inSnow Mountain were collected. The DNA was extracted from specimens bymethod of Chelex-100extraction. The genotypes of15STR loci in Mongolianpopulation in Snow Mountain were generated by multiplexing PCR using theSinofiler PCR system and genotyping using ABI3130xl Genetic Analyzer andthe consequence were analysed by the Genemapper Dv3.3software. Thegenetic parameters of each loci were calculated by Modified-Powerstatessoftware. According to genetic frequency of selected5STR loci, we finallycomputed genetic distances among Mongolian ethnic group in Snow Mountainand other29domestic ethnical groups. And the phylogenetic tree wereanalysed by Neighbour-Joining method of Phylip3.66software.Results224alleles and491genotypes were observed in15STR loci. The allele frequency and genotype frequency were ranged from0.0051~0.4217and0.0051~0.2980, respectively. All loci meet Hardy-Weinberg expectations. Theheterozygosities (Ho) of the15STR loci were between0.7071and0.8737withaverage heterozygosity being0.8007. The polymorphism information content(PIC) was from0.6437to0.8527. The discrimination powers (DP) were from0.6437to0.8527, with the together discrimination power (TDP) beingexceeding0.9999993. The exclusion power (EP) were ranged from0.4393to0.7421with the cumulative exclusion power (CEP) being exceeding0.999999997.The genetic distance among Mongolian population in Snow Mountain andother ethnic groups which closed to it were smaller than that among Mongolianpopulation in Snow Mountain and other ethnic groups which far to it. Theanalysis of phylogenetic tree represent that the genetic relationship betweenMongolian population in Snow Mountain and the Salar nationality is theclosest.ConclusionsThe genetic polymorphism of the15STR loci in Mongolian populationof Gansu province is high. The study data suggest that the13loci of SinofilerSystem (except CSF1PO and D3S1358) can be applied as candidate markersof forensic individual identification and paternity testing. The geneticrelationship between Mongolian population in Snow Mountain and theMongolian population of Qinghai province is the closest.
