ObjectiveTo investigate the effect of Dendrobium mixture on insulin resistance (IR) liver cell and its possible mechanism,in order to provide the theoretical basis for the basic research and clinical application.MethodsLiver cells were incubated with5×10-5mol·L-1insulin for24h. After the model of insulin resistance linver cell were established, incubated them with Dendrobium mixture medicated serum for24hours.Then3.5×10-5mol·L-1insulin stimulation3hours, GLU-GOD-PAP method measuring nutrient solution residual glucose level, and calculating insulin sensitive index; the same method to detect insulin sensitive index after adding PI3K inhibitor—Wortmannin. Western blotting analysiswas used to detect the expression of INS-R、PI3K、T-AKT and P-AKT.ResultsCompared with resistant group, the insulin sensitive index was significantly increased, and expression of TNS-R、PI3K and P-AKT was significantly increased in medicated serum model group. After adding inhibitor both the insulin sensitive index and P-AKT expression were significantly reduce.ConclusionsDendrobium mixture may improve the insulin sensitive index of insulin resistance liver celll,and may be associated with the normal transmission of PI3K/AKT signal pathway.
