| OBJECTIVE:1ã€Explore NOB1(n in one binding protein) mRNA expression inpapillary thyroid carcinoma。2ã€Explore NOB1(n in one binding protein) expression in papillarythyroid carcinoma and its clinical significance。METHOD:Application of real-time reverse transcriptase-polymerase chainreaction (real-time-PCR) detected50cases of primary thyroid papillarycarcinomas and corresponding normal tissue expression of N0B1mRNA,and analysis of the expression and clinical pathologic correlation.Application of immunohistochemical SP method: analysis of50cases of primary thyroid papillary carcinoma,40patients with benignthyroid (20cases of thyroid adenoma and nodular goiter in20cases),10cases of expression of N0B1proteins in normal thyroid, and analysis ofthe expression and clinical pathologic correlation.RESULTS:Real-time PCR results showed: in papillary thyroid carcinoma andthyroid adenoma and nodular goiter is NOB1mRNA expression, which is not expressed in normal thyroid tissue (23425.67-15789.64), NOB1expression in papillary thyroid carcinoma (45678.23-2884.25) issignificantly higher than of thyroid gland (24651.89-13758.72) andnodular goiter (22379.39±14331.67B), statistically significant (p<0.05),however, NOB1in thyroid cancer and normal thyroid tissue, nosignificant difference between Nodular goiter.Immunohistochemical study shows in50cases of papillary thyroidcarcinoma in patients with positive expression in45cases, the positiverate for90%, thyroid adenoma and nodular goiter in normalcircumstances, no tumor cells and stromal fibroblasts immunohistochemicalstaining, proving NOB1mainly on cancer cell localization, ProvingNOB1protein expressions in thyroid cancer groups increasedsignificantly as compared with the other groups (p<0.05), in this study,we analyzed NOB1protein expression and relationship between clinicalpathological variables, including gender, age, stage and the extent oflymph node metastasis in papillary thyroid cancer, what PearsonChi-square (χ2) test. We found NOB1protein expression and UICCstaging as well as there is a clear correlation between tumor size andlymph node metastasis (p<0.05). NOB1expression clearly associatedwith UICC staging (p=0.006). However, NOB1protein expression andno clear relevance of gender and age. CONCLUSION:1ã€mRNA expression level of NOB1was significantly upregulated inthyroid carcinoma tissue, in comparison with thyroid adenoma andnormal thyroid tissues and multinodular goitre tissue by real time PCR.2ã€Protein expression level of NOB1was significantly upregulatedin thyroid carcinoma tissue, in comparison with thyroid adenoma andnormal thyroid tissues and multinodular goitre tissue byimmunohistochemistry.3ã€A high level of NOB1protein expression was associated withlarge tumour size and Union for International Cancer Control stage, butwas not significantly correlated with sex or age, which indicates that theexpression level of the NOB1gene in the thyroid may play a key role inthe occurrence and development of papillary thyroid carcinoma. |
