ObjectivesTo detect the expression of programmed death ligand-1(PD-L1)and interferon-γ(IFN-γ)in papillary thyroid carcinomas and normal thyroid tissue,the relationship between them and clinicopatholigical characteristics was analyzed.To explore the possible role of PD-L1 and IFN-γin papillary thyroid carcinomas,and provide molecular basis for immunotargeting therapy of papillary thyroid carcinomas.MethodsTo collect 29 paraffin-embedded specimens from patients with papillary thyroid carcinomas from November 2017 to July 2018 in the Department of thyroid and breast,Hebei General Hospital.Another 10 cases of contralateral normol thyroid tissue were taken as the control group.The clinical data of the patients were sorted out,including gender,age,tumor size,clinical stage of tumor,violations of thyroid capsule,number of tumor,and lymph node metastasis.The experiment was divided into two groups,including the papillary thyroid carcinoma group(test group)and the normal thyroid tissue group(control group).The location and expression levels of the two molecules were detected by immunohistochemical methods.The amount of expression was determined by analysis with Image Pro Plus 6.0 software.The expression levels of the papillary thyroid carcinoma group were analyzed in combination with clinicopatholigical characteristics.All data were analyzed by SPSS 21.0.Results 1 PD-L1 was mainly localized in the cell membrane or/and cytoplasm.The mean option density of the normal thyroid group and the papillary thyroid carcinoma group were respectively 0.000(0.000,0.001)and 0.007(0.003,0.012).The rank-sum test(Mann-Whitney U test)showed that the expression of PD-L1 protein in the normal thyroid group and papillary thyroid carcinoma group was statistically different(P<0.05).Combined with mean option density,the expression of PD-L1 protein in the papillary thyroid carcinoma group was significantly higher than that in the normal thyroid group.2 The expression of PD-L1was analyzed according to the clinicopatholigical characteristics of papillary thyroid carcinomas:There was no significant correlation between PD-L1 protein expression and gender,age,tumor size,clinical stage of tumor,violations of thyroid capsule,number of tumor,and lymph node metastasis(P>0.05).3 IFN-γwas mainly localized in the cell membrane or/and cytoplasm.The mean option density of the normal thyroid group and the papillarythyroidcarcinomagroupwererespectively0.002(0.000,0.003)and0.013(0.007,0.026).The rank-sum test showed that the expression of IFN-γprotein in the normal thyroid group and papillary thyroid carcinoma group was statistically different(P<0.05).Combined with mean option density,the expression of IFN-γprotein in the papillary thyroid carcinoma group was significantly higher than that in the normal thyroid group.4 The expression of IFN-γwas analyzed according to the clinicopatholigical characteristics of papillary thyroid carcinomas:There was no significant correlation between IFN-γprotein expression and gender,age,tumor size,clinical stage of tumor,violations of thyroid capsule,and number of tumor(P>0.05),but it was correlated with lymph node metastasis(P<0.05).The mean option density of papillary thyroid carcinoma with lymph node metastasis was higher than that of papillary thyroid carcinoma without lymph node metastasis,and there was statistical difference between them.5 The expression of PD-L1 and IFN-γprotein was not correlated in the papillary thyroid carcinoma group.The Spearman correlation coefficient was r_s=0.143,P=0.460,P>0.05.Conclusions1 The expression of PD-L1 and IFN-γin papillary thyroid carcinoma was significantly higher than that in normal thyroid tissue,suggesting that they are associated with the occurrence and progression of papillary thyroid carcinoma.2 The expression of IFN-γin papillary thyroid carcinoma was associated with lymph node metastasis,suggesting that IFN-γmay promote lymph node metastasis of papillary thyroid carcinoma.3 PD-L1 and IFN-γdo not necessarily have a synergistic effect in the development of papillary thyroid carcinoma.Figure2;Table4;Reference 178...
