Background and Objective Observation of insulin-like growth factor I (insulin-like growth factor11GF-]), CD34(marker of tumor vascular endothelial cells, to reflect microvessel density) protein expression in normal colorectal mucosa, adenoma, colorectal carcinoma, And to explore the relationship between the expression and clinicopathological parameters in colorectal cancer IGF-1, CD34, to further explore the relationship with tumor angiogenesis.Methods The expression of IGF-1and CD34was detected. Using immunohistochemical method (two EnVision System), in20cases of nomal tissue,54cases of colorectal adenoma,56cases of colorectal carcinoma. All cases are Proved by pathology. The vascular endothelial cells were marked by CD34, which it counts microvessel (microvessel density MVD).Results1. IGF-1protein was weakly expressed in normal colorectal mucosa. IGF-1protein expression was moderate in colorectal adenoma tissues, and mainly expressed in the cytoplasm. The IGF-1protein expression was higher intensity in colorectal adenocarcinoma, and expressed mainly in the cytoplasm. The positive expression rate of IGF-1in normal colorectal mucosa, colorectal adenoma and colorectal carcinoma were35%,64.81%,85.71%. Comparison of the positive rate of3kinds of tissues, adenoma was higher than that of normal mucosa, adenocarcinoma is higher than that of adenoma, adenocarcinoma was significantly higher than that in normal mucosa. They differences were statistically significant (P<0.05). The expression of IGF-1protein in patients with gender, age, size, site, DUCKE’S staging, there is no thrombi showed no significant correlation (P>0.05). The expression of IGF-1protein in patients with degree of differentiation, depth of tumor invasion, lymph node metastasis were significantly correlated (P<0.05).2,CD34protein expression was low in normal colon tissue, the MVD value was8.7665±2.66732; The CD34protein expression was moderate in colorectal adenoma tissues, the MVD value was22.5320±5.69328; The CD34protein expression was higher in the colorectal carcinoma, the MVD value was35.5534±7.77696. Adenoma group MVD value was significantly higher than that in normal colorectal mucosa, colorectal cancer tissues was significantly higher than that adenoma in group MVD, colorectal cancer tissues was significantly higher than that in normal mucosa group MVD, the groups was statistically significant (<0.05). CD34marker MVD values associated with gender, size, position, there is no thrombi were not significantly correlated (P>0.05), and with age. degree of differentiation, depth of tumor invasion, DUCKE’S staging, lymph node metastasis significantly (P<0.05).Conclusions IGF-1protein are expressed in colorectal adenoma and colorectal adenocarcinoma, Occurrence, development of which may be involved in carcinogenesis of colorectal adenoma, and involved in the regulation of tumor angiogenesis.Expression of MVD and IGF-1strong important indexes can be used as an evaluation node invasion in rectal cancer and malignant degree.
