| Objectives: To observed the influence of Huazhuo Jiedu Fang on the levelsof serum TGF-β1and liver tissue CTGF in hepatic fibrosis rats. To explore theeffect and mechanism of Huazhuo Jiedu Fang to prevent and control the Hepaticfibrosis in rats, in order to provide the theoretical basis for the extensiveapplication of Hua zhuo Jiedu Fang in clinical.Methods: To establish DMN hepatic fibrosis rat model.70grade clean SDrats, male, weight (200+20g) were randomly divided into normal control group(n=10), model control group (n=20), BiejiaruanganPian group (n=10), theHuazhuo Jiedu Fang low, medium and high dose group,10in each group. Afterthe rats were adaptive fed1week, in addition to the normal control group weregiven intraperitoneal injection(1%DMN10mg/kg), a total of13times. At the1st week, the drugs were used continuously3days,1time/day;2-6weeks, thedrugs were used continuously2days,1time/day; The drug delivery begins at9a.m on every Monday. To4th weekend, moderate hepatic fibrosis in rats modelwas built. Dosage: starting from4weeks to eight at the weekend. The normalgroup and the model group were given saline10ml/kg to fill the stomach,1time/day. Huazhuo Jiedu Fang low, medium and high dose group were givenrespectively doses of1.35g/ml,2.7g/ml,5.4g/ml of liquid medicine decoctionto fill the stomach, volume for10ml/kg,1/d. Beijiaruangan Pian group,thetablet grinding into powder,with distilled water mixed suspension, dose of0.154mg/kg,1/d. At the end of8weeks, blood from the abdominal aorta, serum wasisolated. Anatomy and quickly removed the liver, watched the color, texture ofthe liver. Taking about2mm thick liver tissue along the long axis of the liver, and then fixed it in10%Neutral formalin, conventional preparation of liver biopsyand slice thickness of about4mu m. The level of TGF-β1and in liver tissueCTGF in serum was measured with ELISA. The data with mean and standarddeviation (x±s) show, using SPSS13.0statistical software processing.Comparing two sample mean through t test, Comparing multiple sample meanby random design and single factor variance analysis.Results:1The states of rats: The rats in normal control group were eating well, fursmooth and glossy, good mental state, weight growth step by step. The rats inmodel group were eating bad, depressed, dark skin dryness, urine color is yellow,weight increase slightly, but lighter than the normal group. The treatment groupwere eating very well, in good spirits, in good health, weight increase gradually,compared with normal group, the difference was not significant.2The influence of Huazhuo Jiedu Fang on the liver pathologicalmorphology of hepatc fibrosis in rats by visual inspection. The liver texture of therats in normal control group was soft, edge sharp circle, surface smooth, fresh,dark red, no spots, bumps. The liver of rats in model group was big, qualitativehard, dark yellow, rough surface, blunt edge. The liver of rats in treatment groupwas enlargement less obvious, qualitative brittle, slightly dark red, surface smooth,with occasional sand granular hyperplasia. Under the light microscope,thehepatic lobules of rats of normal group were arranged neat set. The fibrous tissuecontent is few. The liver cells of rats in model group degeneration and necrosis,fibrous tissue hyperplasia, structure of hepatic lobule damage. Compared withmodel group, the liver damage is lighter in treatment group. But the differencebetween each treatment group was not obvious in pathology.3. The influence of Huazhuo Jiedu Fang on the levels of serum TGFβ1ofhepatic fibrosis in rats. Compared with the normal group, the levels of serumTGFβ1in model group increased, the difference was statistically significant(P<0.01). Compared with the model group, The levels of serum TGFβ1in thetreatment group reduced. The decreases in Huazhuo Jiedu Fang middle dosegroup was most obvious (P <0.01). 4. The influence of Huazhuo Jiedu Fang on the levels of liver tissue CTGFof hepatic fibrosis in rats. Compared with the normal group, the levels of livertissue CTGF in model group increased, the difference was statistically significant(P<0.01). Compared with the model group, The levels of liver tissue CTGF in thetreatment group reduced. The decreases in Huazhuo Jiedu Fang middle dosegroup was most obvious (P <0.01).Conclusions:1. Huazhuo Jiedu Fang could reduce the degree of liver tissue inflammationand fibrosis development, improve the general condition, showed good effect inprotecting liver and anti-fibrosis in rats.2. Huazhuo Jiedu Fang could significantly reduce the levels of TGFβ1in theserum and liver tissue CTGF of hepatic fibrosis in rats. The effect may be one ofthe mechanism of Huazhuo Jiedu Fang in anti-fibrosis. The activation of HSCwas restrained by reducing the content of TGF-beta1, in liver tissue CTGF,consequently, the synthesis of ECM reduced, the ECM degradation increased,then prevent the excessive accumulation of ECM and help it to play a role inresisting hepatic fibrosis. |
