Lepisorus is a large genus of Polypodiaceae, which is widely distributed in all parts of the country. The Lepisorus plants are rich in flavonoids, phenols, steroids, ecdysterone and other active ingredients. Domestic researches on Lepisorus are mainly in taxonomy, systematics, morphology, anatomy and geographical distribution, but there is few reports about its chemical composition. In this paper, we studied the extraction process of chlorogenic acid, and determined chlorogenic acid, ecdysterone, total flavonoids and antioxidant capacity in vitro in five kinds of Lepisorus plants. In addition, the antioxidant activity in vivo of the ethanol extraction of rhizome in Lepisorus marginatus was studied. The main results are as follows:(1) The effects of the extraction solvent concentration, ratio of solvent to sample, extraction time and ultrasonic power were evaluated by single factor analysis and the orthogonal analysis.The optimal conditions of solvent extraction were obtained as follows:ethanol concentration70%, ratio of solvent to sample1:30, extraction time30min and ultrasonic power150W. Under this condition, the extraction volume of chlorogenic acid is2.57%. By HPLC the chlorogenic acid and ecdysterone contents in five kinds of Lepisorus plants were compared, the results show the chlorogenic acid and ecdysterone contents are different in the five kinds of plants, and the contents in Lepisorus marginatus and Lepisorus macrosphaerus are relatively higher. As for the same species, the contents in the rhizome part are higher than the vegetative leaves. In addition, by using UV-spectrophotometry the total flavonoids were determined and compared in five kinds of Lepisorus plants, and the results show different species and different medicinal parts of the same kind of plant are different in the content of the total flavonoids.(2) We adopted capbility of removing DPPH free radical, inhibit β-carotene/linoleic acid system, capbility of removing hydroxyl radical and capability of deoxidizing as the indexes to prove the five kinds of Lepisorus plants have strong anti-oxidative capability in vitro. The results show the alcohol extracts of the five kinds of Lepisorus plant and different medicinal parts have strong radical scavenging activity towards DPPH system and hydroxyl radical syetem, and also significant in reducing power and β-carotene/linoleic acid oxidation inhibiting activity.(3) By making the comprehensive analysis of antioxidant activity compounds and the oxidation resistance in vitro among the five kinds of Lepisorus plants, we used the ethanol extraction of rhizome in common Lepisorus marginatus in qinling mountain area as the fluid, determined the levels of T-AOC, T-SOD and MDA in liver tissue and blood serum of mice, and studied the antioxidation effect in vivo of the rhizome from Lepisorus marginatus. The results show that the ethanol extraction of Lepisorus marginatus’s rhizome could improve the activity of T-AOC and T-SOD and remarkably reduce MDA content in serum and liver tissue of mice,which indicated the ethanol extraction of rhizome in Lepisorus marginatus can improve antioxidation ability in vivo and balance the state of antioxidation/oxidation.
