Morphological changes, apoptotic cells and expression of VEGF were observed by H&E staining, TUNEL in situ labeling and VEGF immunohistochemical staining. Statistical analyses were conducted to confirm these results.Results:1. H&E staining:Obvious morphological changes were observed in submandibular and parotid glands under light microscopy. The results showed the submandibular and parotid glands atrophy and the significant morphological changes in acinar and duct. Irregular arrangement of gland cells were clustered accumulation. It was showed a certain degree of recovery that the acinar and duct atrophy extent become alleviate in the submandibular gland and parotid of Xueshuantong treatment groups compared with diabetic group.2. Apoptotic cells in Tunel:â‘ submandibular gland:The percentage of apoptotic cells was significantly increased in diabetic group than that in control group (P<0.05) and Xueshuantong treatment groups (P<0.05). There is no significant difference between normal group and Xueshuantong treatment groups (p>0.05).â‘¡parotid gland:The percentage of apoptotic cells was significantly increased in diabetic group than that in control group (P<0.05) and Xueshuantong treatment groups (P<0.05) while significantly decreased in normal group than that in Xueshuantong treatment groups (P<0.05). Apoptotic cells of small dose treatment group were more than large dose treatment group (p>0.05).3. VEGF immunohistochemical staining:â‘ submandibular gland:The expression percentage of VEGF was significantly increased in diabetic group than that in other three groups (P<0.05). The normal group got significantly decreased VEGF expression compared to the Xueshuantong treatment groups (P<0.05) while no significant difference between the two treatment groups (P>0.05).â‘¡parotid gland:The expression percentage of VEGF was significantly increased in diabetic group than that in other three groups (P<0.05). The normal group got significantly increased VEGF expression compared to the small dose treatment group (P <0.05) while decreased compared to the large dose treatment group (P<0.05).The small dose treatment group got significant decreased VEGF expression than the large dose treatment group (P<0.05).Conclusion:1. The atrophy of submandibular and parotid glands of diabetic rats was more severe than the normal group and treatment groups.2. The percentage apoptotic cells and VEGF expression of diabetic rats were more than the normal group and treatment groups.3. Xueshuantong could decrease the number of apoptotic cells of diabetic rats, especially in submandibular gland. Xueshuantong could improve the VEGF expression of diabetic rats.
